目的:探讨Smac在食管癌组织中的表达及其与Caspase-3之间的关系.方法:采用免疫组织化学S-P法检测Smac和Caspase-3在食管癌组织和癌旁正常食管组织中的表达情况.结果:Smac在食管癌旁正常食管组织中的表达明显高于癌组织( P<0.05).Smac表达与癌组织分化程度无关( P>0.05).Caspase-3在癌旁正常食管组织中表达明显高于癌组织( P<0.05).在食管癌组织Smac与Caspase-3表达呈正相关( P<0.05).结论:Smac在食管癌组织中异常表达,有望成为食管癌诊断和基因治疗的新靶点.Smac基因与凋亡相关基因Caspase-3的异常表达可能在食管癌癌变中起协调作用.

Objective:To study the expression of Smac and its relationship with expression of Caspase-3 in esophageal carcinoma. Methods:Immunohistochemical S-P method was used to detect the expression of Smac and Caspase-3 proteins in 40 esophageal carcinoma tissues and 20 para-carcinoma tissues. Results:Smac protein was expressed in 23 of 40 esophageal carcinoma tissues (57.5%),but Smac protein showed high level in para–cancerous tissues(90.0%, P<0. 05).However, Smac protein expression was not associated with the different histological grade ( P>0. 05). Caspase-3 protein was expressed in 21 of 40 (52.5% ) esophageal carcinoma tissues, but Caspase-3 protein showed high level in para–cancerous tissues(85.0%, P<0. 05). Smac protein expression was positively related to Caspase-3 protein expression in esophageal carcinoma tissues ( P<0. 05). Conclusion:Aberrant expression of Smac may be a new target for diagnosis and gene treatment of esophageal carcinoma. The aberrant expression of Smac and apoptosis related gene Caspase-3 may play synergetic roles in process of carcinogenesis of esophageal carcinoma.
