目的:观察依那普利对百草枯(PQ)中毒大鼠肺组织病理改变、血管紧张素转化酶(ACE)表达的影响。方法一次性PQ 60 mg/kg灌胃建立大鼠百草枯中毒肺纤维化模型,干预组染毒后予依那普利10 mg/(kg?d)灌胃。于染毒后3 d、7 d、14 d取肺组织行HE染色和Masson染色,观察大鼠肺泡炎、肺纤维化程度;Quantitative Real-Time PCR检测ACE mRNA表达。结果干预组在染毒后3d、7d肺泡炎症减轻;染毒7d、14d后,纤维化程度积分低于模型组。模型组染毒后各时间点大鼠肺组织ACE mRNA表达逐步上调,持续至14 d。干预组各时间点ACE mRNA低于模型组。结论依那普利可能通过降低局部ACE表达,从而减轻百草枯中毒所致肺纤维化形成。

목적:관찰의나보리대백초고(PQ)중독대서폐조직병리개변、혈관긴장소전화매(ACE)표체적영향。방법일차성PQ 60 mg/kg관위건립대서백초고중독폐섬유화모형,간예조염독후여의나보리10 mg/(kg?d)관위。우염독후3 d、7 d、14 d취폐조직행HE염색화Masson염색,관찰대서폐포염、폐섬유화정도;Quantitative Real-Time PCR검측ACE mRNA표체。결과간예조재염독후3d、7d폐포염증감경;염독7d、14d후,섬유화정도적분저우모형조。모형조염독후각시간점대서폐조직ACE mRNA표체축보상조,지속지14 d。간예조각시간점ACE mRNA저우모형조。결론의나보리가능통과강저국부ACE표체,종이감경백초고중독소치폐섬유화형성。
Objective To investigate the effects of enalapril on histopathologic alteration and the expression of angiotensin converting enzyme (ACE) gene in lung of paraquat (PQ) poisoning rats. Methods A rat model of pulmonary ifbrosis was induced by intragastric administration with a single dose (60 mg/kg) of paraquat. Then intervention group was treated intragastrically with enalapril at a dose of 10 mg/(kg?d). Rats were sacriifced and lungs were harvested at different time of day 3, day 7 or day 14 following poisoning. The level of pulmonary al-veolitis and ifbrosis were estimated by using HE staining and Masson staining. The expression of ACE mRNA was measured by quantitative Real-Time PCR. Results Compared with model group, intervention group exhibited signs of less serious pulmonary alveolitis on day 3 and day 7. Similarly, intervention group brought a lower ifbro-sis scores on day 7 and day 14. After PQ exposure, lung ACE mRNA expression observed in model group were gradually up-regulated at each time point and were maintained until day 14. Expression levels of ACE mRNA in lung tissue of intervention group were lower than those of model group at all time points. Conclusion Enalapril could alleviate paraquat-induced pulmonary ifbrosis, which may be due to suppressing ACE mRNA expression in local tissue.
