目的:探讨CXCR4和VEGF- C在胃癌淋巴结转移中的作用及两者的相关性。方法应用免疫组化法及RT- PCR检测CXCR4及VEGF- C在人胃癌组织中的表达水平,并分析其与胃癌淋巴结转移的关系。结果 CXCR4及VEGF- C蛋白在癌旁组织中无阳性表达,在胃癌组织中均呈阳性表达。胃癌组织中CXCR4及VEGF- C蛋白阳性表达率均明显高于癌旁组织(P<0.05或0.01),且N0组织、N1+N2组织、N3组织中CXCR4及VEGF- C蛋白阳性表达率两两比较差异均有统计学意义(P<0.05或0.01)。不同胃癌组织中CXCR4mRNA及VEGF- CmRNA的表达明显不同,差异均有统计学意义(P<0.05或0.01)。胃癌组织中CXCR4表达与VEGF- C表达呈显著正相关(r=0.355,P<0.01)。结论胃癌中CXCR4和VEGF- C的表达呈正相关,两者的协同作用与胃癌患者淋巴结转移密切相关,进一步研究两者的作用机制可能对预测淋巴结转移及判断预后有重要临床价值。

목적:탐토CXCR4화VEGF- C재위암림파결전이중적작용급량자적상관성。방법응용면역조화법급RT- PCR검측CXCR4급VEGF- C재인위암조직중적표체수평,병분석기여위암림파결전이적관계。결과 CXCR4급VEGF- C단백재암방조직중무양성표체,재위암조직중균정양성표체。위암조직중CXCR4급VEGF- C단백양성표체솔균명현고우암방조직(P<0.05혹0.01),차N0조직、N1+N2조직、N3조직중CXCR4급VEGF- C단백양성표체솔량량비교차이균유통계학의의(P<0.05혹0.01)。불동위암조직중CXCR4mRNA급VEGF- CmRNA적표체명현불동,차이균유통계학의의(P<0.05혹0.01)。위암조직중CXCR4표체여VEGF- C표체정현저정상관(r=0.355,P<0.01)。결론위암중CXCR4화VEGF- C적표체정정상관,량자적협동작용여위암환자림파결전이밀절상관,진일보연구량자적작용궤제가능대예측림파결전이급판단예후유중요림상개치。
Objective To investigate the association of CXCR4 and VEGF- C with lymphatic metastasis in gastric cancer. Methods The expressions of CXCR4 and VEGF- C proteins and mRNAs in human gastric cancer were detected by immunohisto-chemistry and RT- PCR, respectively. The relationship between CXCR4 and VEGF- C expressions and lymph node metastasis of gastric cancer was analyzed. Results CXCR4 and VEGF- C proteins were positively expressed in gastric cancer tissue, while they were not detected in pericancerous gastric tissue. The positive rate of CXCR4 and VEGF- C protein expression in gastric cancer tissue was significantly higher than that in pericancerous gastric tissue(P<0.05 or 0.01). There were significant differences in positive expression rates of CXCR4 and VEGF- C proteins and mRNAs among stage N0, N1+N2 and N3 gastric cancers (P<0.05或0.01). The expression level of CXCR4 was positively correlated with that of VEGF- C (r=0.355, P<0.01). Conclusion The synergy of CXCR4 and VEGF- C was closely related to the lymphatic metastasis of gastric cancer, and further study on the mechanism may has important clinical value in predicting lymph node metastasis and prognosis.
