目的 观察微小RNA(miR)-155在乳腺癌中的表达及对乳腺癌HBL-100细胞株迁移能力的影响.方法 通过实时定量逆转录聚合酶链反应(RT-qPCR)法检测60例正常乳腺组织及60例乳腺癌组织中miR-155的表达水平,分析其与临床病理特征的关系.同时在乳腺癌HBL-100细胞株中分别转染miR-155-mimics及抑制物,采用Transwell小室实验检测细胞的迁移能力.结果 miR-155在乳腺癌中的表达明显高于正常乳腺组织(P<0.01),其表达与淋巴结转移及TNM分期明显相关(P<0.05).Transwell实验检测结果表明miR-155过表达后,细胞的迁移数为(46.3±4.6)个,明显高于对照组[(31.6±2.2)个,P<0.05];而miR-155表达下调后,细胞的迁移数明显减少,为(20.0±1.9)个(P<0.05).结论 miR-155表达与乳腺癌的形成密切相关,并影响细胞的迁移能力.

Objective To investigate the expression of microRNA (miR)-155 in breast cancer and its effect on the migration in HBL-100 cell line.Methods Real-time quantitative reverse polymerase chain reaction (RT-qPCR) was used to detect the expression of miR-155 mRNA in 60 cases of breast cancer tissues and 60 cases of normal breast tissues.The relationship between the miR-155 expression and clinicopathological characteristics was analyzed in breast cancer.Tfter transfection of miR-155 mimics and inhibitor into HBL-100 cells respectively,the ability of migration was evaluated by Transwell assay.Results The miR-155 expression in breast cancer tissues was significantly higher than that in normal breast tissues (P <0.01).The results of Transwell assay showed that the number of migrating cells in high miR-155 expression group was 46.3 ±4.6,significantly greater than that in control group (31.6 ±2.2) (P <0.05).After miR-155 down-regulation,the number of migrating cells was strongly reduced (20.0 ± 1.9) (P <0.05).Conclusion miR-155 expression is closely related to the formation of breast cancer,and also affects cell migration.
