目的:探讨孕鼠暴露低剂量邻苯二甲酸二己酯( di-2-ethylhexylphthalate ,DEHP)对仔鼠脂肪分布的影响及机制。方法:将C57BL/6J孕鼠分为2组,对照组喂饲基础饲料,DEHP组喂饲添加质量分数为0.06%DEHP的基础饲料,至仔鼠出生。9周后,处死仔鼠,切取附睾周围和腹股沟皮下脂肪组织,称质量;实时定量RT-PCR法检测Gpc4、Tbx15 mRNA表达。结果:DEHP组仔鼠附睾脂肪组织质量显著高于对照组( P<0.05);对照组和DEHP组仔鼠附睾脂肪组织Gpc4 mRNA表达均显著高于皮下脂肪组织( P<0.05),而Tbx15 mRNA表达则显著低于皮下脂肪组织( P<0.01);相比对照组, DEHP组仔鼠附睾脂肪组织 Gpc4 mRNA及皮下脂肪组织Tbx15 mRNA表达均显著增加( P<0.01)。结论:胚胎期低剂量DEHP暴露可能通过改变内脏及皮下Gpc4和Tbx15的表达,影响脂肪组织的分布。

목적:탐토잉서폭로저제량린분이갑산이기지( di-2-ethylhexylphthalate ,DEHP)대자서지방분포적영향급궤제。방법:장C57BL/6J잉서분위2조,대조조위사기출사료,DEHP조위사첨가질량분수위0.06%DEHP적기출사료,지자서출생。9주후,처사자서,절취부고주위화복고구피하지방조직,칭질량;실시정량RT-PCR법검측Gpc4、Tbx15 mRNA표체。결과:DEHP조자서부고지방조직질량현저고우대조조( P<0.05);대조조화DEHP조자서부고지방조직Gpc4 mRNA표체균현저고우피하지방조직( P<0.05),이Tbx15 mRNA표체칙현저저우피하지방조직( P<0.01);상비대조조, DEHP조자서부고지방조직 Gpc4 mRNA급피하지방조직Tbx15 mRNA표체균현저증가( P<0.01)。결론:배태기저제량DEHP폭로가능통과개변내장급피하Gpc4화Tbx15적표체,영향지방조직적분포。
Objective: To investigate the effect and mechanism of low dose di-2-ethylhexylphthalate ( DEHP ) exposure on fat distribution of offspring in pregnant mice .Methods: Pregnant C57BL/6J mice were divided into two groups.Mice in control group were fed with chow diet , and mice in DEHP group were fed with chow diet supplemented with 0.06%( w/w ) DEHP till their offspring was born .Nine weeks after birth , the mice were sacrificed .Epididymal and inguinal subcutaneous adipose tissues were dissected and weighted .The expression of Gpc4 and Tbx15 mRNA was measured by real-time reverse transcription polymerase chain reaction ( RT-PCR ) method.Results:The weight of epididymal adipose tissues in DEHP group was significantly higher than that in control group (P<0.05).The expression of Gpc4 mRNA in epididymal adipose tissues was significantly higher than that in subcutaneous adipose tissues ( P<0.05 ) , while the expression of Tbx 15 mRNA was significantly lower than that in subcutaneous adipose tissues ( P<0.01 ) in both control and DEHP group .Compared with control group , Gpc4 mRNA expression in epididymal adipose tissues and Tbx 15 mRNA expression in subcutaneous adipose tissues were increased ( P<0.01) .Conclusion:Low dose DEHP exposure in mice prenatal period perhaps affects fat distribution via modifying the expression of Gpc 4 and Tbx15 mRNA in visceral and subcutaneous adipose tissues.
