目的::为了探讨与人血细胞相关的GATA1转录因子在实体肿瘤发生发展中的功能,构建GATA1的慢病毒干扰载体,并验证其敲低效果。方法:根据人GATA1的cDNA序列,设计含有小发卡结构的寡核苷酸序列,将其克隆到慢病毒表达载体上;将重组质粒转染人胚肾293T细胞,通过实时定量RT-PCR及Western印迹检测GATA1的表达水平。结果和结论:构建了具有明显干扰效果的GATA1基因的小干扰RNA(siRNA)慢病毒表达载体,能够有效抑制GATA1 mRNA和蛋白水平,为后续的GATA1生物学作用研究奠定了基础。

Objective: To study the biological function of human blood cells-associated GATA1 in the development and progression of solid tumor, the lentiviral vector for GATA1 small-interfering RNA(siRNA) was constructed and its knockdown effect was determined. Methods: GATA1 siRNA was designed and constructed based on human GATA1 cDNA sequence using a lentiviral vector. The expression of GATA1 was examined by real-time RT-PCR and Western blot analysis. Results & Conclusion: A lentivirus RNA interference vector targeting GATA1 gene was successfully constructed, which laid the foundation for further study of GATA1 function.
