采用生物信息学方法对GenBank, SwissProt中的拟南芥、大豆、甘蓝型油菜、小麦等物种的乙酰辅酶A羧化酶核苷酸和氨基酸序列进行了比对分析,进而对其组成成分、理化性质、疏水性/亲水性、跨膜结构域、蛋白质二级结构、功能结构域、三级结构数进行了预测和分析.结果表明: ACC基因具有完整的开放阅读框,长约50683 bp,编码氨基酸相对分子质量为93.41 kD,理论等电点为7.27,是亲水性不稳定蛋白,二级结构均以α-螺旋和不规则卷曲为主要构件.通过进化树分析可分别将8种生物分为2组: A支包括小麦等7个物种,B支仅含甘蓝型油菜.

The nucleic acid sequence and amino acid sequence of acetyl-CoA carboxylase (ACC) from Ara-bidopsis thaliana, Glycine max, Brassica napus, and Triticum aestivum, which were registered in GenBank and SwissProt, were analyzed and predicted with the tools of bioinformatics in the following main aspects, i.e. the composition of nucleic acid sequence and amino acid sequence, physical and chemical characters, hydrophobicity or hydrophilicity, transmembrane topologieal structure, secondary structure of protein, functional domains and tertiary structure. The results showed that ACC gene contained a complete open reading frame with length of 50 683 bp, the molecular weight of amino acids was 93.41 kD, the theoretical isoelectric point was 7.27. This was a hydrophilic and instable protein, in which alpha helix and random coil were the main components of all secondary structures. The eight species were divided into two groups by phylogenetic analysis, A branch including seven species including Triticum aestivum, and B branch only containing B. napus.
