目的:探讨骨髓间充质干细胞( BMSC )修复兔破损硬脊膜的可行性和有效性。方法穿刺抽取日本大耳白兔髂后上棘骨髓,分离BMSC,采用贴壁筛选法对分离的BMSC进行纯化,接种于人工硬脊膜上。16只日本大耳白兔,随机分成A组、B组,每组8只,制作L4椎板切除硬脊膜破损模型, A组人工硬脊膜敷贴裂口处, B组骨髓间充质干细复合人工硬脊膜敷贴裂口处。术后30天处死动物,取修复区域的组织行大体观察和病理学观察,并对成纤维细胞密度进行分析。结果 B组成纤维细胞与胶原纤维丰富,细胞密度高于A组( P<0.05)。结论骨髓间充质干细胞可以促进硬脊膜修复。

목적:탐토골수간충질간세포( BMSC )수복토파손경척막적가행성화유효성。방법천자추취일본대이백토가후상극골수,분리BMSC,채용첩벽사선법대분리적BMSC진행순화,접충우인공경척막상。16지일본대이백토,수궤분성A조、B조,매조8지,제작L4추판절제경척막파손모형, A조인공경척막부첩렬구처, B조골수간충질간세복합인공경척막부첩렬구처。술후30천처사동물,취수복구역적조직행대체관찰화병이학관찰,병대성섬유세포밀도진행분석。결과 B조성섬유세포여효원섬유봉부,세포밀도고우A조( P<0.05)。결론골수간충질간세포가이촉진경척막수복。
Objective To explore the feasibility and effectiveness of bone marrow mesenchymal stem cells ( BM-SCs) in repair of defects in rabbit dura mater .Methods The bone marrow was extracted from the ilium of rabbits . Then, BMSCs were isolated and purified through by density gradient centrifugation and adherence screening method before seeded onto artificial spinal dura mater .Sixteen rabbits were randomly divided into group A and group B , with eight in each group.An epidural damage model was made by removal of the L 4 vertebral plate.Then, artificial spinal dura mater was padded into the gap in group A , while artificial spinal dura mater combined with bone marrow mesenchymal stem cells was padded into the gap in group B .The animals were sacrificed 30 days after operation and the tissue within the re-paired area was collected for gross pathological and histological observation .Cell density was also calculated .Results Large amounts of fibroblasts and collagen fibers were observed in group B .Cell density in group B was higher than that of group A (P<0.05).Conclusion Bone marrow mesenchymal stem cells can promote the restoration of the dura mater .
