目的:探索碱性成纤维细胞生长因子(bFGF)-壳聚糖载体对成年大鼠骨髓间充质干细胞(rMSCs)向神经细胞转化的诱导作用。方法采用免疫组织化学进行定性观察,并定量计数rMSCs诱导转化为神经干细胞、神经元和星形胶质细胞等神经细胞,噻唑蓝(MTT)比色法定量检测诱导后细胞的活性。结果 rMSCs经bFGF-壳聚糖载体诱导后,表达神经干细胞标记物nestin、神经元标记物β-tubulinⅢ和星形胶质细胞标记物胶质纤维酸性蛋白(GFAP), nestin在bFGF-壳聚糖组中高表达,9 d时达到最大值49.40%。结论 bFGF-壳聚糖载体可以诱导成年rMSCs高比例地分化为神经干细胞。

목적:탐색감성성섬유세포생장인자(bFGF)-각취당재체대성년대서골수간충질간세포(rMSCs)향신경세포전화적유도작용。방법채용면역조직화학진행정성관찰,병정량계수rMSCs유도전화위신경간세포、신경원화성형효질세포등신경세포,새서람(MTT)비색법정량검측유도후세포적활성。결과 rMSCs경bFGF-각취당재체유도후,표체신경간세포표기물nestin、신경원표기물β-tubulinⅢ화성형효질세포표기물효질섬유산성단백(GFAP), nestin재bFGF-각취당조중고표체,9 d시체도최대치49.40%。결론 bFGF-각취당재체가이유도성년rMSCs고비례지분화위신경간세포。
Objective To explore the induction of basic fibroblast growth factor (bFGF)-chitosan carrier transforming the adult rat bone marrow mesenchymal stem cells (rMSCs) into nerve cells. Methods rMSCs were detected qualitatively and counted quantitatively by immu-nohistochemistry after they were induced into nerve cells, such as neural stem cells neurons and astrocytes. The methyl thiazolyl tetrazolium (MTT) chromometry assay was carried out to determine the cell viability. Results rMSCs induced by bFGF-chitosan carrier expressed neu-ral stem cell marker nestin, neuron markerβ-tubulinⅢand astrocytes marker glial fibrillary acidic protein (GFAP). Nestin expressed more in the bFGF-chitosan group, and reached its maximum (49.40%) at the 9th day. Conclusion bFGF-chitosan carrier can induce the adult rM-SCs differentiate into neural stem cells in a high proportion.
