银杏黄酮糖苷因具有多种生物活性而被制药、食品、日用品等诸多领域广泛应用。针对目前银杏黄酮糖苷检测操作繁琐、成本高、耗时长的不足,建立了一种高精确、低成本的快速测定方法。根据黄酮类化合物能与A l3+反应形成荧光螯合物的原理,以芦丁为标样探索测试条件,结果表明在激发波长λex =400 nm ,发射波长λem =520 nm ,pH为3.6的Al(NO3)3-(HAc-NaAc)反应体系中反应1500 s ,螯合物荧光强度趋于稳定且达到最大值。求得芦丁浓度与荧光值的线性回归方程为 y=29.92 x+36.49(R2=0.986),线性范围为1.8×10-6~3.2×10-5 mol·L -1。用这种方法检测了银杏悬浮培养细胞中黄酮糖苷的含量,并进行加标回收实验,平均回收率为101.3%。该方法灵敏度高,重现性好,操作简单,具有良好的应用前景。

Flavonoids glycosides from ginkgo are widely applied in pharmaceutical ,food ,daily necessities and other areas due to their variety of biological activities .To overcome the shortcoming of current methods of time wasting ,complex operation and expensive costs ,an accuracy ,low costs and fast determination method was established for flavonoids glycosides from ginkgo . According to the principle that flavonoids can form fluorescent chelate with Al 3+ ,the detection condition was explored with rutin as standard .The results showed that the fluorimetric intensy of chelate of rutin with Al 3+ would be stabilized and achieve maxi-mum with λex =400 nm and λem =520 nm ,in Al(NO3 )3-(HAc-NaAc) reaction system for 1 500 s with pH 3.6 .The linear re-gression equation y=29.92x+36.49 (R2 =0.986) was deduced with the concentration of rutin and fluorescence intensy ,and the linear range 1.8 × 10-6 ~3.2 × 10-5 mol · L -1 .Flavonoids glycosides of cell suspension cultures from ginkgo biloba was detec-ted by this method .The recovery experiments were also carried out with the average recovery rate of 101.3% .The advantages of high sensitivity ,reproducibility ,simple operation ,and low costs were showed ,indicating its good prospects .
