目的:探讨氯硝柳胺及EDTA对不同螺龄湖北钉螺酚氧化酶(PO)活性的影响。方法将不同螺龄湖北钉螺(即成螺、幼螺、螺卵)软体匀浆、离心获取粗制酶液,再依次通过盐析、透析及凝胶过滤层析而获取部分纯化的酚氧化酶酶液。以邻苯二酚作为底物,检测不同浓度氯硝柳胺及EDTA对不同螺龄湖北钉螺PO活性的影响。结果随着氯硝柳胺和EDTA浓度的增大,不同螺龄湖北钉螺PO活性均逐渐降低。结论氯硝柳胺和EDTA对不同螺龄湖北钉螺PO活性均有抑制作用。

목적:탐토록초류알급EDTA대불동라령호북정라분양화매(PO)활성적영향。방법장불동라령호북정라(즉성라、유라、라란)연체균장、리심획취조제매액,재의차통과염석、투석급응효과려층석이획취부분순화적분양화매매액。이린분이분작위저물,검측불동농도록초류알급EDTA대불동라령호북정라PO활성적영향。결과수착록초류알화EDTA농도적증대,불동라령호북정라PO활성균축점강저。결론록초류알화EDTA대불동라령호북정라PO활성균유억제작용。
Objective To investigate the effects of niclosamide and EDTA on the activity of phenoloxidase(PO) from Oncomelania hupensis in different ages. Methods The soft tissues from Oncomelania hupensis in different ages, such as adult snails, young snails and the eggs of snails, were homogenated and centrifugated to obtain crude enzyme liquid, then purified by salt precipitation, dialysis and gel filtration to obtain PO. The activity of PO was detected with catechol as the substrate in the condition of different concentrations of niclosamide and EDTA. Results The activity of PO from Oncomelania hupensis in different ages was decreasing with the concentration elevating of niclosamide and EDTA. Conclusion Both niclosamide and EDTA have inhibition effect on the activity of PO from Oncomelania hupensis in different ages.
