采用海藻酸钠-壳聚糖固定化碱性果胶酶,通过对固定化酶活力回收率影响条件的讨论,确定碱性果胶酶固定化的最优条件为:海藻酸钠3%,碳酸钙1.5%,游离酶1500 U/g,乳化时间50 min,乳化剂MOA-3用量0.4 mL/L,转速500 r/min,壳聚糖1.5%。结果表明:固定化酶活力回收率达到69.23%。制备的固定化酶最适pH为9,最适温度为60℃,这种固定化酶比游离酶的耐热性和耐碱性有明显提高,而且固定化操作稳定性较好,重复利用5次后,酶活力仍可保持原来的60%。
採用海藻痠鈉-殼聚糖固定化堿性果膠酶,通過對固定化酶活力迴收率影響條件的討論,確定堿性果膠酶固定化的最優條件為:海藻痠鈉3%,碳痠鈣1.5%,遊離酶1500 U/g,乳化時間50 min,乳化劑MOA-3用量0.4 mL/L,轉速500 r/min,殼聚糖1.5%。結果錶明:固定化酶活力迴收率達到69.23%。製備的固定化酶最適pH為9,最適溫度為60℃,這種固定化酶比遊離酶的耐熱性和耐堿性有明顯提高,而且固定化操作穩定性較好,重複利用5次後,酶活力仍可保持原來的60%。
채용해조산납-각취당고정화감성과효매,통과대고정화매활력회수솔영향조건적토론,학정감성과효매고정화적최우조건위:해조산납3%,탄산개1.5%,유리매1500 U/g,유화시간50 min,유화제MOA-3용량0.4 mL/L,전속500 r/min,각취당1.5%。결과표명:고정화매활력회수솔체도69.23%。제비적고정화매최괄pH위9,최괄온도위60℃,저충고정화매비유리매적내열성화내감성유명현제고,이차고정화조작은정성교호,중복이용5차후,매활력잉가보지원래적60%。
Alkaline pectinase was immobilized by sodium-chitosan alginate. With the discussion of the influencing of activity recovery rate factors, the optimal conditions for immobilization of alkaline pectinase were confirmed as follows:the concentration of sodium alginate 3%, concentration of calcium carbonate 1.5%, amount of free enzyme 1 500 U/g, emulsification time 50 min, amount of emulsifier MOA-3 0.4 mL/L, speed of emulsification 500 r/min, chitosan alginate 1.5%. It is found that the activity recovery rate of immobilized enzyme can reach 69.23%. The optimum application conditions for the prepared immobilized enzyme were pH 9 at 60℃ Compared to free enzyme, the thermal resistance and alkali resistance were obviously improved. Furthermore, the operational stability was better. Being used for five cycles, the enzyme activity can still maintain 60%that of the original′s.
