目的 探讨癌症高表达蛋白1(HEC1)在结直肠癌组织中的表达及其临床意义.方法 选取本院40例手术切除癌组织的结直肠癌患者作为研究对象,收集标本采用免疫组织化学及实时荧光定量聚合酶链反应(FQ-PCR)两种方法测定结直肠癌组织中的HEC1的表达,并分析其与结直肠癌患者的临床病理特征的相关性.结果 免疫组织化学结果显示HEC1在结直肠癌组织中的阳性表达率为70%,明显高于癌旁结直肠组织的15%(P<0.05);结直肠癌组织中HEC1的表达与癌症的临床病理分期以及有无转移明显相关.40例结直肠癌组织中HEC1 mRNA表达量为24±4,明显高于40例癌旁结直肠组织中的HEC1 mRNA表达量为9±5,未见HEC1基因突变.结论 HEC1过度表达在结直肠癌的发生发展中发挥着重要的作用,考虑与非整倍体细胞数量增加有关,结合临床分析HEC1的检测利于对结直肠患者进行诊治并判断预后.

Objective To investigate the expression of ancer high expression protein 1 (HEC1) in colorectal cancer tissues and its clinical significance.Methods Forty cases of colorectal cancer subject to surgical resection were selected.The expression of HEC1 in colorectal cancer tissues was detected by immunohistochemistry and real-time fluorescent quantitative polymerase chain reaction (FQ-PCR),and its correlation with clinicopathological characteristics of colorectal cancer patients was analyzed.Results The immunohistochemistry revealed that the positive expression rate of HEC1 in colorectal cancer tissues was 70%,significantly higher than in the normal colorectal tissues adjacent to carcinoma (15%,P < 0.05).The HEC1 expression in colorectal cancer tissues was significantly associated with clinicopathologic staging and metastasis.CT value showed that the HEC1 expression in 40 cases of colorectal cancer tissues was 24 ± 4,significantly higher than in 40 cases of normal colorectal tissues adjacent to carcinoma (9 ± 5).No HEC1 gene mutations were detected.Conclusion Overexpression of HEC1 may play important roles in the pathogenesis of colorectal cancer.The combined detection of HEC1 expression clinical may be beneficial for the diagnosis,treatment and prognosis evaluation of patients with colorectal cancer.
