目的:探讨白介素-6(IL-6)及基质金属蛋白酶-2(MMP-2)在胚胎发育中的可能作用。方法:对30例不明原因胚胎停止发育病例(研究组)和30例正常早孕吸宫流产病例(对照组)的绒毛组织及血清进行研究。采用实时荧光定量逆转录聚合酶链式反应技术(Real time-PCR)和免疫组织化学技术(SP 法)检测两组绒毛组织中 IL-6和 MMP-2 mRNA 的表达和蛋白定位、表达强度;采用双抗体酶联免疫夹心法(ELISA法)测定IL-6在两组血清中的浓度。结果:绒毛组织中,IL-6与MMP-2在合体及绒毛滋养层细胞中均有表达,Real time-PCR 及免疫组化结果均显示IL-6在研究组中的表达水平低于对照组,MMP-2在研究组中的表达水平高于对照组,差异均有统计学意义(P<0.05); IL-6在血清中的表达水平组间无统计学差异(P>0.05)。结论:孕早期绒毛组织中IL-6和MMP-2的正常表达对维持早期胚胎发育起到一定作用。

목적:탐토백개소-6(IL-6)급기질금속단백매-2(MMP-2)재배태발육중적가능작용。방법:대30례불명원인배태정지발육병례(연구조)화30례정상조잉흡궁유산병례(대조조)적융모조직급혈청진행연구。채용실시형광정량역전록취합매련식반응기술(Real time-PCR)화면역조직화학기술(SP 법)검측량조융모조직중 IL-6화 MMP-2 mRNA 적표체화단백정위、표체강도;채용쌍항체매련면역협심법(ELISA법)측정IL-6재량조혈청중적농도。결과:융모조직중,IL-6여MMP-2재합체급융모자양층세포중균유표체,Real time-PCR 급면역조화결과균현시IL-6재연구조중적표체수평저우대조조,MMP-2재연구조중적표체수평고우대조조,차이균유통계학의의(P<0.05); IL-6재혈청중적표체수평조간무통계학차이(P>0.05)。결론:잉조기융모조직중IL-6화MMP-2적정상표체대유지조기배태발육기도일정작용。
Objective To explore the relationship between the Interleukin 6 , matrix metalloproteinases 2 and early embryo arrest. Methods Real time-PCR was used to measure the mRNA expression of IL-6 and MMP-2 and immunohistochemistry (IHC, SP method)was used to measure the location and expression of the two different kinds of protein in villus. ELISA was used to measure the level of IL-6 in serum. Results Real-time PCR and IHC showed that the expression levels of IL-6 was significantly lower in experimental group than in control group, and MMP-2 was significantly higher than the control group (P < 0.05). Differenc of IL-6 level in serum between the two groups was not statistically significant (P > 0.05). Conclusion Proper expressions of IL-6 and MMP-2 in the villus tissue play a key role in the maintenance of early pregnancy.
