采用外部换热中心移热式反应器合成了环形整体固定相基质,对基质进行化学改性制备了弱阴离子(DEAE)整体色谱柱,分别采用健合及吸附法将牛血清白蛋白(BSA)固载到基质上,制备适用于快速分离当归有效成分的径向亲和整体柱,并考察了整体色谱柱的分离性能。结果表明吸附牛血清白蛋白(BSA)色谱柱的分离性能优于健合蛋白色谱柱。

In this paper, a porous polymer monolith was prepared in a thermostated annular reactor. Weak anion ion-exchange radial flow monolithic columns were prepared by further surface modification of the stationary phase. Bovine serum albumin(BSA)was immobilized on the matrix, by means of adsorbed and bonded proteins respectively. The prepared radial flow affinity monolithic columns were then used to separate pharmaceutical constituents of Angelica. The separation efficiency of these monolithic columns was also investigated. It was shown that the affinity column prepared by adsorption of BSA achieved higher separation efficiency than the one with bonded BSA.
