目的观察在体大鼠心肌缺血的不同再灌注时程NF-κBp65与AngⅡ表达的变化。方法40只大鼠采用随机数字表法分为R15min组、R30min组、R60min组、R120min组及对照组。经缺血30min及不同时间的再灌注(15,30,60,120 min)而建立在体心肌缺血再灌注损伤模型。以ELISA法定量检测NF-κBp65蛋白的DNA结合活性;采用放射免疫分析法检测血浆及心肌组织 AngⅡ含量。通过 Pearson相关性检验对 NF-κBp65与循环及心肌局部的 AngⅡ含量做相关性分析。结果NF-κBp65蛋白表达量随再灌注时间的延长呈升高趋势,于R60min时达高峰,R120min组与R60min组比有所下降,差异均有统计学意义(均P<0.05),但仍高于R30min组;各实验组血浆及心肌局部AngⅡ含量与对照组比明显升高,随再灌注时间的延长呈升高趋势,差异有统计学意义(P<0.05),两者呈直线正相关(r=0.859、0.809,均P<0.05)。结论心肌缺血再灌注损伤时NF-κBp65及血液与心肌AngⅡ均增高。

목적관찰재체대서심기결혈적불동재관주시정NF-κBp65여AngⅡ표체적변화。방법40지대서채용수궤수자표법분위R15min조、R30min조、R60min조、R120min조급대조조。경결혈30min급불동시간적재관주(15,30,60,120 min)이건립재체심기결혈재관주손상모형。이ELISA법정량검측NF-κBp65단백적DNA결합활성;채용방사면역분석법검측혈장급심기조직 AngⅡ함량。통과 Pearson상관성검험대 NF-κBp65여순배급심기국부적 AngⅡ함량주상관성분석。결과NF-κBp65단백표체량수재관주시간적연장정승고추세,우R60min시체고봉,R120min조여R60min조비유소하강,차이균유통계학의의(균P<0.05),단잉고우R30min조;각실험조혈장급심기국부AngⅡ함량여대조조비명현승고,수재관주시간적연장정승고추세,차이유통계학의의(P<0.05),량자정직선정상관(r=0.859、0.809,균P<0.05)。결론심기결혈재관주손상시NF-κBp65급혈액여심기AngⅡ균증고。
Objective To observe the changes of NF-κB p65 and AngⅡ during various periods of myocardial ischemia-reperfusion. Methods 40 rats were randomly divided into R15min, R30min, R60min and R120min groups and control group. Rat model of myocardial ischemia-reperfusion was established based on 30-min ischemia fol owed by reperfusion for various periods (15, 30, 60, 120min). The DNA-binding activity of NF-κB p65 was detected quantitatively by ELISA. AngⅡin blood plasma and myocardium were measured by radioimmunoassay. The relevance of NF-κB p65 to AngⅡin blood plasma and myocardium was analyzed by Pearson correlation test. Results The expression of NF-κB p65 had a tendency toward increase in paral el to the prolongation of reperfusion, with the highest in group R60min, fol owed by group R120min and group R30min. There was significantly different between any two groups (P<0.05). AngⅡ in blood plasma and myocardium was significantly higher in each ischemia-reperfusion group than in control group, which gradual y increased with the prolongation of reperfusion (P<0.05). There was a positive linear correlation between NF-κB p65 and Ang II in blood plasma and myocardium(r=0.859、0.809, P<0.05). Conclusion NF-κB p65 as wel as AngII in blood plasma and myocardium increase during myocardial ischemia-reperfusion injury.
