目的:探讨miRNA-625*在膀胱尿路上皮癌中的差异表达。方法常规提取新鲜膀胱尿路上皮癌与正常膀胱黏膜组织中总RNA;使用分光光度计测定RNA在260 nm、280 nm和230 nm的吸光值,以计算浓度并评估纯度;用琼脂糖凝胶电泳检测RNA纯度及完整性;以RT-PCR方法检测标本中miRNA-625*的表达情况。结果 RNA质量检测合格后, miRNA-625*经RT-PCR验证与miRNA芯片分析结果一致。与正常膀胱黏膜上皮组织相比,膀胱尿路上皮癌中miRNA-625*表达显著上调( P<0.05)。结论 miRNA-625*在膀胱尿路上皮癌中表达上调,可作为膀胱尿路上皮癌诊断和预后判断的分子标志物。

목적:탐토miRNA-625*재방광뇨로상피암중적차이표체。방법상규제취신선방광뇨로상피암여정상방광점막조직중총RNA;사용분광광도계측정RNA재260 nm、280 nm화230 nm적흡광치,이계산농도병평고순도;용경지당응효전영검측RNA순도급완정성;이RT-PCR방법검측표본중miRNA-625*적표체정황。결과 RNA질량검측합격후, miRNA-625*경RT-PCR험증여miRNA심편분석결과일치。여정상방광점막상피조직상비,방광뇨로상피암중miRNA-625*표체현저상조( P<0.05)。결론 miRNA-625*재방광뇨로상피암중표체상조,가작위방광뇨로상피암진단화예후판단적분자표지물。
Objective To detect the miRNA-625*expression in urothelial carcinoma of the bladder ( BUC) .Methods BUC and normal bladder mucosa tissues were collected and the total RNA was extracted routinely .Measuring the absorption value of RNA at 260 nm,280 nm,and 230 nm by NanoDrop ND-1000 in order to calculate RNA Quantification and Quality Assurance .RNA Integrity and gDNA contamination Test by Denaturing Agarose Gel Electrophoresis .The miRNA-625*expression was detected by using real-time quantitative polymerase chain reaction .Results RNA Quantification and RNA Integrity were qualified for the follow-up experiment ,the miRNA-625* finding was confirmed by real-time RT-PCR and the expression was significantly up-regulated in BUC tissues than those in normal bladder mucosa (P<0.05).Conclusion The expression level of miRNA-625*is significantly up-regulated in BUC,and it may be used for prediction of prognosis and as for targeted treatment of BUC.
