目的:通过仿生法在多孔钛支架表面构建含有牛血清白蛋白的钙磷涂层,评价牛血清白的整合对钙磷涂层形貌、化学成分的影响。方法:利用选择性激光熔覆方法制备多孔钛支架,将多孔钛支架经碱液处理后,浸入模拟体液中4d进行预钙化,随后将2组支架分别浸泡于含牛血清白蛋白浓度为10μg/mL的饱和钙磷溶液、不含牛血清白蛋白的饱和钙磷溶液中2d。检测支架表面涂层形貌及物相组成。结果:牛血清白蛋白加入后,钙磷涂层晶体尺寸变小,由微米级结构变为纳米级;涂层成分由羟基磷灰石和磷酸八钙混合相转变为碳酸化的羟基磷灰石。结论:成功在多孔钛支架表面构建了牛血清白蛋白/钙磷复合涂层;牛血清白蛋白整合到钙磷涂层内部。
目的:通過倣生法在多孔鈦支架錶麵構建含有牛血清白蛋白的鈣燐塗層,評價牛血清白的整閤對鈣燐塗層形貌、化學成分的影響。方法:利用選擇性激光鎔覆方法製備多孔鈦支架,將多孔鈦支架經堿液處理後,浸入模擬體液中4d進行預鈣化,隨後將2組支架分彆浸泡于含牛血清白蛋白濃度為10μg/mL的飽和鈣燐溶液、不含牛血清白蛋白的飽和鈣燐溶液中2d。檢測支架錶麵塗層形貌及物相組成。結果:牛血清白蛋白加入後,鈣燐塗層晶體呎吋變小,由微米級結構變為納米級;塗層成分由羥基燐灰石和燐痠八鈣混閤相轉變為碳痠化的羥基燐灰石。結論:成功在多孔鈦支架錶麵構建瞭牛血清白蛋白/鈣燐複閤塗層;牛血清白蛋白整閤到鈣燐塗層內部。
목적:통과방생법재다공태지가표면구건함유우혈청백단백적개린도층,평개우혈청백적정합대개린도층형모、화학성분적영향。방법:이용선택성격광용복방법제비다공태지가,장다공태지가경감액처리후,침입모의체액중4d진행예개화,수후장2조지가분별침포우함우혈청백단백농도위10μg/mL적포화개린용액、불함우혈청백단백적포화개린용액중2d。검측지가표면도층형모급물상조성。결과:우혈청백단백가입후,개린도층정체척촌변소,유미미급결구변위납미급;도층성분유간기린회석화린산팔개혼합상전변위탄산화적간기린회석。결론:성공재다공태지가표면구건료우혈청백단백/개린복합도층;우혈청백단백정합도개린도층내부。
Objectives: To fabricate apatite coating containing bovine serum albumin by co-deposition method on the surface of porous titanium scaffolds, and evaluate effects of incorporation of bovine serum albumin (BSA) on the morphology and chemical composition of the bioactive coating. Methods:The porous titanium scaffolds were prepared by using selective laser melting method. After alkali treatment, the porous titanium scaffolds were immersed into simulated body fluid for 4days for pre-calcification, subsequently two groups scaffolds were soaked in saturated calcium phosphate solution without or with containing BSA concentration of 10 μg/mL for 2 days. The topography of the specimen was observed by scanning electron microscopy (SEM). Chemical structure and phase composition of coatings were detected by Fourier infrared spectroscopy (FTIR) analysis and X-ray diffraction (XRD) respectively. Results: the addition of bovine serum albumin reduced crystal size of calcium phosphate, changing from micron grade structure to nanoscale;and coating without BSA was composed of hydroxyapatite and octocalcium phosphate, while BSA/calcium phosphate coatings contained hydroxyapatite. Conclusion: We successfully fabricated hybrid coating of BSA/calcium phosphate coatings on porous titanium scaffolds, and crystal size of coating reduced with the addition of BSA .
