为了探明玉米高赖氨酸奥帕克-2(opaque-2,o2)和奥帕克-16(opaque-16,o16)突变的分子遗传差异,以太系19/o2为o2突变基因供体亲本,QCL3021和 QCL3024为o16突变基因的供体亲本,普通系QCL2179和 QCL2180为受体轮回亲本,构建o2和o16近等基因系(NIL),经过多代回交和自交,并利用SSR分子标记技术。选育近等基因系,并对其进行遗传背景分析。结果表明:多代回交和自交后选育出BC5 F2世代的o2和o16近等基因系(NIL)25份;从40个SSR标记中,筛选出25个在亲本间具有多态性的SSR标记;25个系基于分子标记的遗传背景恢复率为90%~100%,仅有7个系遗传背景恢复率高于理论值(98.4%),表明仍需继续进行回交选择。

In order to conduct molecular genetic research on maize high lysine mutants of opaque-2 (o2 )and opaque-1 6 (o16 ).The high lysine genes o2 and o16 were backcrossed into normal lines QCL2 1 7 9 and QCL2180 using the maize high lysine inbreds Taixi19 (o2),and QCL3021(o16)and QCL3024(o16)as donors and normal maize inbred lines QCL2179 and QCL2180 as receptors.Twenty five BC5F2 near-isogenic lines (NILs)of o2 and o16 were bred through multiple backcross and self-cross.Twenty five polymorphic SSR markers between parents among 40 SSR markers were selected to detect genetic background of the NILs.Results:The genetic background recovery rate of 25 NILs was from 90% to 100% based on molecular markers.However,only 7 NILs had higher rate than theoretical value (98.4%), suggesting that the NILs still need continuous backcross selection.
