目的:建立维药睡莲花中没食子酸和烟花苷含量的高效液相色谱测定方法。方法色谱柱:Wondasil C18(250 mm×4.6 mm,5μm);流动相:A为甲醇,B为0.1%冰乙酸混合水溶液,梯度洗脱;流速:1.0 mL/min;检测波长:270 nm;柱温:30℃。结果没食子酸及烟花苷进样量分别在0.065~0.585μg(r=0.9991)、0.133~1.197μg(r=0.9995)范围内与峰面积呈良好的线性关系,平均回收率分别为102.71%、102.08%,RSD分别为1.97%、0.46%。结论本方法操作简单、准确可靠,可用于维药睡莲花中烟花苷和没食子酸含量的测定。

Objective To establish an HPLC method for the content determination of gallic acid and kaempferol-3-O-rutinoside inNymphaea candida Presl..MethodsThe separation was carried out on a Wondasil C18 column (250 mm×4.6 mm, 5μm). The mobile phase was methanol-0.1% acetic acid solution, with gradient elution;the flow rate was 1.0 mL/min;the detection wavelength was set at 270 nm;the column temperature was 30℃.Results The linear ranges of gallic acid and kaempferol-3-O-rutinoside were 0.065-0.585μg (r=0.999 1), 0.133-1.197μg (r=0.999 5), respectively. The average recoveries of gallic acid and kaempferol-3-O-rutinoside were 102.71%, 102.08%, with RSD of 1.97%, 0.46%, respectively.Conclusion This method was rapid, accurate, and reliable.
