目的:观察清心安神方含药血清对豚鼠离体心室肌细胞内向钾电流(Ik1)。方法:分离豚鼠的心室肌细胞,采用全细胞膜片钳技术电流钳模式记录动作电位,观察含40%的空白血清及含药血清的细胞外液灌流对 Ik1的影响。结果:清心安神方含药血清对豚鼠心室肌细胞内向钾电流(Ik1)影响:40%含药血清组对IKl内向电流成分影响为:40%含药血清组从(-30.68±10.39)pA/pF抑制为(-21.03±3.47)pA/pF(n=7 P<0.05)。结论:清心安神方含药血清对大鼠心室肌细胞IKl内向成份有抑制作用,呈浓度依赖性。

목적:관찰청심안신방함약혈청대돈서리체심실기세포내향갑전류(Ik1)。방법:분리돈서적심실기세포,채용전세포막편겸기술전류겸모식기록동작전위,관찰함40%적공백혈청급함약혈청적세포외액관류대 Ik1적영향。결과:청심안신방함약혈청대돈서심실기세포내향갑전류(Ik1)영향:40%함약혈청조대IKl내향전류성분영향위:40%함약혈청조종(-30.68±10.39)pA/pF억제위(-21.03±3.47)pA/pF(n=7 P<0.05)。결론:청심안신방함약혈청대대서심실기세포IKl내향성빈유억제작용,정농도의뢰성。
Objective:To observe the influence of serum containing Heart-Clearing and Spirit-Calming Decoction on inward rectifier potassium channel current in ventricular myocytes isolated from Rat. Methods:Record the action potential by whole patch clamp technique and currentc lamp m ode. Observe the r espective influence of extracellular fluid perfusion of 40%blank serum and drug serum on IK1. Results:At the test with voltage of-100mV,the inward component of IK1 in the 40%drug group was decreased from(-30.68±10.39)pA/pF to(-21.03±3.47)pA/pF(n=7 P<0.001). Conclusion:40%serum containing Heart-Clearing and Spirit-Calming Deooction can decrease the IK1. It was concentration dependent.
