本文的研究内容是奶牛性控精子膜蛋白双向电泳平台的建立和优化以及性控X、Y精子膜蛋白表达的差异性分析。分别对性控X精子、Y精子提取膜蛋白并进行双向电泳,用ImageMaster7.0软件对本结果进行比对分析,发现一张图谱上大约有600左右的蛋白点,其中差异蛋白点32个,在差异蛋白点中,上调蛋白19个,下调蛋白13个。说明基于双向电泳的蛋白质组学可以用于性控精子的研究,发生改变的这些差异蛋白可能在性别分化过程中发挥重要作用。

The objective of this study was to extract the sexed sperm membrane protein of dairy cattle and conduct two-dimensional gel electrophoresis. The protein of cattle spermatozoa was separated by two-dimensional gel electrophoresis technique, comparative analysis of the sexed semen membrane protein was carried out by the ImageMaster7.0. The results revealed that there are about 600 protein spots in each map, 32 spots were different, among them 19 spots were up and 13 spots were fall. This indicates that two-dimensional gel electrophoresis can used for sexed sperm study, the proteins that are differentially regulated may present potential biomarkers of sex differentiation.
