目的:采用高效液相色谱法-蒸发光散射检测法测定痔速宁片中猪去氧胆酸和鹅去氧胆酸的含量。方法采用Thermo U-3000液相色谱系统;ChromeleonTM 7工作站;ELSD 2000ES检测器,检测器条件:漂移管温度:105℃,撞击管关闭,增益为1,气体流速:2.0 L/min。采用Thermo(4.6×150 mm,C185μm 120 A)色谱柱;流动相:乙腈-0.1%冰醋酸(50:50);流速:0.9 mL/min;柱温:20℃。结果表明猪去氧胆酸在(21.530~262.884)μg/mL(r=0.9997),鹅去氧胆酸在(11.148~133.776)μg/mL(r=0.9995)的范围内呈良好的线性关系;猪去氧胆酸平均加样回收率为101.8%(n=9),RSD=2.8%;鹅去氧胆酸平均加样回收率为98.0%(n=9),RSD=2.3%。结论本方法操作简便、准确、灵敏度高,可用于痔速宁片的质量控制。

Objective To determine the hyodeoxycholic acid and chenodeoxycholic acid in zhisuning tablet by HPLC-ELSD. Methods Used Thermo U-3000 HPLC, ChromeleonTM 7 and ELSD 2000ES, drift tube temperature was 105℃, impactor was off, gain was 1, gas flow was 2.0 L/min;Thermo column (4.6í150 mm,C18 5μm 120A) was used with the methanol-0.1%Glacial acetic acid (50:50) as mobile phase at a flow rate of 0.9 mL/min; The column temperature was 20℃. Results The linear range of hyodeoxycholic acid was (21.530~262.884) μg/mL(r=0.9997), the average recovery was 101.8%(n=9), RSD=2.8%; The linear range of chenodeoxycholic was (11.148~133.776) μg/mL(r=0.9995), the aver-age recovery was 98.0%(n=9), RSD=2.3%. Concluslon This method is simple, sensitive, and accurate for the quality control of astraloside in zhisuning tablet.
