为提高微杆菌Z4发酵的几丁质酶得率,利用摇瓶研究了发酵温度、初始p H、装液量等对于发酵产酶的影响.在这些实验的基础上,运用正交试验对Z4产几丁质酶的摇瓶发酵条件进行了进一步优化.结果的极差分析和方差分析表明温度对产酶的影响最大,具有显著性.最佳摇瓶发酵工艺条件为:培养温度28℃,培养基初始p H 7,接种量6%,三角瓶装液量60 m L培养基/250 m L三角瓶.在该条件下发酵液酶产量达到4.13 U/m L ,比优化前提高了79.6%.
為提高微桿菌Z4髮酵的幾丁質酶得率,利用搖瓶研究瞭髮酵溫度、初始p H、裝液量等對于髮酵產酶的影響.在這些實驗的基礎上,運用正交試驗對Z4產幾丁質酶的搖瓶髮酵條件進行瞭進一步優化.結果的極差分析和方差分析錶明溫度對產酶的影響最大,具有顯著性.最佳搖瓶髮酵工藝條件為:培養溫度28℃,培養基初始p H 7,接種量6%,三角瓶裝液量60 m L培養基/250 m L三角瓶.在該條件下髮酵液酶產量達到4.13 U/m L ,比優化前提高瞭79.6%.

The effect of the fermentation temperature ,inoculum ratio and liquid volume of the flask on the chitinase fermentation of Microbacterium sp Z4 was studied .The fermentation condition for optimized production of chitinase was further studied by using L9 (34 ) orthogo-nal experimental method .The maximal chitinase activity up to 4 .13 U/mL was attained un-der the condition :temperature of 28 degrees Celsius ,initial pH 7 ,inoculums 6% ,and 60 mL medium in 250 mL flask .The chitinase production increased 79 .6% of the initial production .
