研究了固定化乳酸脱氢酶( LDH)的制备和催化性质,讨论了戊二醛体积分数、酶的偶联时间、pH值对酶固定化的影响.对游离酶和固定化酶催化性质的研究结果表明:酶促反应最适pH分别为9.2和10.2,最适温度分别为51℃和50℃,米氏常数分别为16.2 mmol/L和0.7 mmol/L.与游离酶相比,固定化酶的活力稳定性更佳,有更好的贮存稳定性和复用性.
研究瞭固定化乳痠脫氫酶( LDH)的製備和催化性質,討論瞭戊二醛體積分數、酶的偶聯時間、pH值對酶固定化的影響.對遊離酶和固定化酶催化性質的研究結果錶明:酶促反應最適pH分彆為9.2和10.2,最適溫度分彆為51℃和50℃,米氏常數分彆為16.2 mmol/L和0.7 mmol/L.與遊離酶相比,固定化酶的活力穩定性更佳,有更好的貯存穩定性和複用性.

In this paper,the preparation and enzymatic characteristics of immobilized lactate dehydrogenase ( LDH) was studied and the impact of glutaraldehyde concentration , enzyme binding time and pH value on immobilization were discussed.The results of the study on the enzymatic characteristics of free and immobilized enzymes showed that the optimum pH value for the enzymatic reaction were 9.2 and 10.2,the optimum temperatures were 51℃ and 50℃,the Km value were 16.2 mmol/L and 0.7 mmol/L respectively.It was shown that the immobilized enzyme has better activity stability, storage stability and reusability .
