为了解天然食物中主要存在的2种共轭亚油酸异构体(c 9,t11-CLA 和 t10,c 12-CLA)改善胰岛素抵抗的活性,建立胰岛素抵抗3T3-L1脂肪细胞模型,分别以5,10,20μmol·L-1的 c 9,t11-CLA 或 t10,c 12-CLA 进行干预处理,测定干预处理后培养液中葡萄糖浓度和细胞中脂肪含量的变化。通过考察空白组(CON)、模型组(MOD)和罗格列酮阳性对照组(ROS),结果发现 c 9,t11-CLA 不能增加胰岛素抵抗脂肪细胞的葡萄糖消耗量,而 t10,c 12-CLA能增加胰岛素抵抗脂肪细胞的葡萄糖消耗量,并且 t10,c 12-CLA 的作用浓度和时间与葡萄糖的消耗量呈一定的量效关系。20μmol·L-1 t10,c 12-CLA 处理组的葡萄糖消耗量和 ROS 相当(P >0.05),但前者细胞中的脂肪含量显著低于后者(P <0.05)。显示 t10,c 12-CLA 改善胰岛素抵抗3T3-L1脂肪细胞的活性可能与其降低细胞中的脂肪含量有关。
為瞭解天然食物中主要存在的2種共軛亞油痠異構體(c 9,t11-CLA 和 t10,c 12-CLA)改善胰島素牴抗的活性,建立胰島素牴抗3T3-L1脂肪細胞模型,分彆以5,10,20μmol·L-1的 c 9,t11-CLA 或 t10,c 12-CLA 進行榦預處理,測定榦預處理後培養液中葡萄糖濃度和細胞中脂肪含量的變化。通過攷察空白組(CON)、模型組(MOD)和囉格列酮暘性對照組(ROS),結果髮現 c 9,t11-CLA 不能增加胰島素牴抗脂肪細胞的葡萄糖消耗量,而 t10,c 12-CLA能增加胰島素牴抗脂肪細胞的葡萄糖消耗量,併且 t10,c 12-CLA 的作用濃度和時間與葡萄糖的消耗量呈一定的量效關繫。20μmol·L-1 t10,c 12-CLA 處理組的葡萄糖消耗量和 ROS 相噹(P >0.05),但前者細胞中的脂肪含量顯著低于後者(P <0.05)。顯示 t10,c 12-CLA 改善胰島素牴抗3T3-L1脂肪細胞的活性可能與其降低細胞中的脂肪含量有關。
위료해천연식물중주요존재적2충공액아유산이구체(c 9,t11-CLA 화 t10,c 12-CLA)개선이도소저항적활성,건립이도소저항3T3-L1지방세포모형,분별이5,10,20μmol·L-1적 c 9,t11-CLA 혹 t10,c 12-CLA 진행간예처리,측정간예처리후배양액중포도당농도화세포중지방함량적변화。통과고찰공백조(CON)、모형조(MOD)화라격렬동양성대조조(ROS),결과발현 c 9,t11-CLA 불능증가이도소저항지방세포적포도당소모량,이 t10,c 12-CLA능증가이도소저항지방세포적포도당소모량,병차 t10,c 12-CLA 적작용농도화시간여포도당적소모량정일정적량효관계。20μmol·L-1 t10,c 12-CLA 처리조적포도당소모량화 ROS 상당(P >0.05),단전자세포중적지방함량현저저우후자(P <0.05)。현시 t10,c 12-CLA 개선이도소저항3T3-L1지방세포적활성가능여기강저세포중적지방함량유관。
Two main Conjugated Linoleic Acids (CLAs)c9,t11 and t10,c12 in natural food were selected to investigate their effects on enhancing insulin sensitivity.The insulin-resistant 3T3-L1 adipocytes were in-duced by high concentration insulin,which was used as cell model.Glucose consumption was tested each 24 h for all treatments.It was found that c9,t11-CLA did not have any activity in enhancing insulin sensitivi-ty.However,t10,c12-CLA showed significant ability to enhance glucose consumption in a concentration and time dependent manner.For instance,at the concentration of 20 μmol·L-1 ,t10,c12-CLA had the same effect as rosiglitazone(P >0.05),which was a commonly used medicine in type-2 diabetes.The content of lipid in the cells was significantly decreased by the treatment of t10,c12-CLA (P <0.05),which might be associated with the activity of enhancing insulin sensitivity.
