目的:探讨川穹嗪对于Aβ1-42致痴呆模型小鼠学习记忆功能的影响,并对其机制进行初步研究。方法:通过避暗实验、跳台实验检测小鼠学习记忆功能;通过胞内钙影像测定胞内钙浓度变化。结果:避暗实验和跳台实验均表明,高中低剂量组的川穹嗪与模型组相比,能显著增加潜伏期,减少错误次数(P<0.05);胞内钙影像结果表明,在有钙或无钙外液两种情况下,川穹嗪均降低海马神经元胞内钙离子浓度。结论:川穹嗪可显著改善Aβ1-42致痴呆模型小鼠学习记忆功能,其机制可能与川穹嗪降低胞内钙离子浓度,从而调节神经元兴奋性,突触可塑性和信号传递有关。

목적:탐토천궁진대우Aβ1-42치치태모형소서학습기억공능적영향,병대기궤제진행초보연구。방법:통과피암실험、도태실험검측소서학습기억공능;통과포내개영상측정포내개농도변화。결과:피암실험화도태실험균표명,고중저제량조적천궁진여모형조상비,능현저증가잠복기,감소착오차수(P<0.05);포내개영상결과표명,재유개혹무개외액량충정황하,천궁진균강저해마신경원포내개리자농도。결론:천궁진가현저개선Aβ1-42치치태모형소서학습기억공능,기궤제가능여천궁진강저포내개리자농도,종이조절신경원흥강성,돌촉가소성화신호전체유관。
Objectives:To discuss tetramethylpyrazine (TMP)’s influence on learning and memorial function of Aβ1-42 caused AD model mouse and its potential mechanism. Methods:Tested mouse’s learning and memorial function through step-through test and step-down test;Measured calcium concentration through intracellular calcium imaging. Results:Behavior test showed that compared to model group, TMP can increase the latent period and reduce error times significantly(P<0.05);calcium image results showed that TMP can reduce calcium concentration of hippocampal neurons in the extracellular solution with calcium or without calcium. Conclusion:TMP can improve learning and memorial functions of Aβ1-42 caused AD mouse significantly. The mechanism may due to TMP can reduce intracellular calcium ion concentration, therefore regulate neuronal excitability, neuronal plasticity and signal transmission.
