目的:利用代谢组学手段检测大脑中动脉阻塞(Middle Cerebral Artery Occlusion,MCAO)模型大鼠血清中氨基酸含量,微观上阐明氨基酸注射液的疗效以及部分作用机制。方法:采用邻苯二甲醛(O-Phthalaldehyde,OPA)柱前在线衍生反相高效液相色谱法,采用荧光检测,梯度洗脱,建立了血清中氨基酸代谢谱的相对定量分析方法,测定36例大鼠血清中的15种氨基酸含量。流动相A为10 mol/L Na2HP04-Na2B07缓冲液(pH=7.95),B为乙腈-甲醇-水(45∶45∶15,V/V/V),线性梯度洗脱,流动相B在6 min内由5%增加至17.2%,6~35 min内由17.2%增加至52%。结果:1)该法在35 min之内分离出15种氨基酸;2)经主成分分析,3个组在空间上得到很好地分离,氨基酸代谢谱发生了一定的变化;3)利用SPSS方差分析,有6个氨基酸在分离各组有统计学意义。结论:丹红注射液通过部分调节MCAO摄动通路改善脑梗死大鼠血清部分氨基酸的代谢趋势。

목적:이용대사조학수단검측대뇌중동맥조새(Middle Cerebral Artery Occlusion,MCAO)모형대서혈청중안기산함량,미관상천명안기산주사액적료효이급부분작용궤제。방법:채용린분이갑철(O-Phthalaldehyde,OPA)주전재선연생반상고효액상색보법,채용형광검측,제도세탈,건립료혈청중안기산대사보적상대정량분석방법,측정36례대서혈청중적15충안기산함량。류동상A위10 mol/L Na2HP04-Na2B07완충액(pH=7.95),B위을정-갑순-수(45∶45∶15,V/V/V),선성제도세탈,류동상B재6 min내유5%증가지17.2%,6~35 min내유17.2%증가지52%。결과:1)해법재35 min지내분리출15충안기산;2)경주성분분석,3개조재공간상득도흔호지분리,안기산대사보발생료일정적변화;3)이용SPSS방차분석,유6개안기산재분리각조유통계학의의。결론:단홍주사액통과부분조절MCAO섭동통로개선뇌경사대서혈청부분안기산적대사추세。
Objective:To determine the content of amino acid to clarify the curative effect and the functioning mechanism of DanHong injection on microscopic .Methods:OPA reversed phase high performance liquid chromatography ( HPLC ) , precolumn derivatization method fluorescence detection, gradient elution were used.Mobile phase A was 15mol/L Na2HP04-Na2B07 buffer (pH =7.95), B was acetonitrile -methanol -water (45:45:15, V/V/V).As for linear gradient elution, mobile phase B in 6 min increased from 5%to 17. 2%, increased from 17.2%to 52%in 6-35 min.Results:1) Fifteen kinds of amino acids were isolated within 35 min;2) using princi-pal component analysis , three groups were well separated;3) using the SPSS analysis of variance , there were six amino acids in the sep-aration between groups was statistically significant .Conclusion:Danhong injection can improve metabolic trend of amino acid in cerebral infarction rats serum through adjusting perturbed pathways.
