目的:探讨信号传导与转录激活因子3(stat3)及细胞周期蛋白D1(cyclinD1)在口腔鳞癌组织中的表达及其与临床预后的关系。方法采用逆转录聚合酶链反应法(RT- PCR)对35例口腔鳞癌患者的癌组织、相应正常组织中stat3、cyclinD1基因mRNA的表达进行检测,并结合临床资料进行分析。结果鳞癌组织中stat3及cyclinD1mRNA的表达水平均明显高于相应正常组织的表达,差异有统计学意义(P<0.05或0.01)。鳞癌组织中stat3mRNA的表达与cyclinD1mRNA的表达无明显相关(P>0.05)。stat3、cyclinD1 mRNA的表达与肿瘤临床分期、淋巴结转移明显相关(P<0.05),而与性别、年龄、发生部位、肿瘤大小和病理分级之间无明显相关(均P>0.05)。结论检测stat3、cyclinD1mRNA的表达可能有助于口腔鳞癌淋巴结转移和肿瘤临床分期的预测;口腔鳞癌发生、发展中stat3基因在mRNA水平上对cyclinD1的调控作用不明显。

목적:탐토신호전도여전록격활인자3(stat3)급세포주기단백D1(cyclinD1)재구강린암조직중적표체급기여림상예후적관계。방법채용역전록취합매련반응법(RT- PCR)대35례구강린암환자적암조직、상응정상조직중stat3、cyclinD1기인mRNA적표체진행검측,병결합림상자료진행분석。결과린암조직중stat3급cyclinD1mRNA적표체수평균명현고우상응정상조직적표체,차이유통계학의의(P<0.05혹0.01)。린암조직중stat3mRNA적표체여cyclinD1mRNA적표체무명현상관(P>0.05)。stat3、cyclinD1 mRNA적표체여종류림상분기、림파결전이명현상관(P<0.05),이여성별、년령、발생부위、종류대소화병리분급지간무명현상관(균P>0.05)。결론검측stat3、cyclinD1mRNA적표체가능유조우구강린암림파결전이화종류림상분기적예측;구강린암발생、발전중stat3기인재mRNA수평상대cyclinD1적조공작용불명현。
Objective To determine the expression and clinical significance of stat3/cyclinD1 pathway in oral squamous cellcarcinoma(OSCC). Methods Tissue specimens of carcinoma and matched normal mucosa were taken from 35 patients with OSCC. The mRNA expression of stat3 and cyclinD1 in tissue specimens were determined by reverse transcription polymerase chain reaction (RT- PCR). Results The mRNA expression of stat3 and cyclinD1 gene in OSCC were significantly higher than that in normal mucosa (P<0.05). The expression of stat3 and cyclinD1 mRNA in OSCC was correlated with lymph node metastasis and clinical stages of tumor (P<0.05), while was not correlated with sex, age, tumor location, tumor size and pathological grade (P>0.05). There was no correlation between expression of stat3 and cyclinD1 mRNA in OSCC tissue specimens (P>0.05). Conclusion Determination of stat3 and cyclinD1 gene expression may contribute to predict lymph node metastasis and clinical stage of oral squaremous cell carcinoma. The results indicate that stat3 gene may not involve in regulation of cyclinD1 expression in the development of OSCC.
