为研究去甲斑蝥素(Norcantharidin,NCTD)对白血病细胞的DR5表达及肿瘤坏死因子相关的凋亡诱导配体(TRAIL)诱导凋亡敏感性的影响,通过流式细胞术检测NCTD和rhTRAIL作用下白血病细胞K562的凋亡、DR5表达和细胞内活性氧(reactive oxygen species,ROS)水平;通过免疫印迹技术分析激酶JNK1的表达。结果表明:NCTD以浓度依赖方式上调 DR5表达;与 rhTRAIL单独作用比较,低浓度的 NCTD(7.5μmol/L)与rhTRAIL共同处理可显著提高K562细胞凋亡百分率、ROS水平和JNK1表达。NCTD通过ROS/JNK途径上调DR5表达,增强了白血病细胞对rhTRAIL的敏感性。

To explore the effects of NCTD on DR5 expression and cell apoptosis in leukaemia cells in-duced by TRAIL,apoptosis,expression of DR5 and level of ROS of leukaemia cells K562 induced by NCTD and rhTRAIL were detected using flow cytometry,and protein kinase JNK1 was measured by west-ern blot assay .The results showed that the expression of DR5 was up-regulated by NCTD in a dose-de-pendent model .In contrast to cells induced by rhTRAIL alone,apoptosis rate,ROS and JNK1 in K562 cells treated by rhTRAIL combined with lower concentration of NCTD (7.5μmol/L)were enhanced signif-icantly.In conclusion,NCTD could sensitize leukaemia cells to rhTRAIL by up-regulation of DR5 through ROS/JNK signaling pathway .
