目的研究碱性成纤维细胞生长因子( bFGF)对山梨醇脱氢酶( SDH)在大鼠系膜细胞( GMCS)及大鼠抗Thy-1肾小球肾炎模型( ATG)中表达的影响。方法采用RT-qPCR和Western blot分别检测bFGF刺激后体外培养GMCS SDH的表达以及bFGF和SDH在ATG中的表达;应用免疫组化检测ATG中bFGF、 SDH的表达,并对染色强度进行图像分析。结果原代培养的GMCs为成纤维细胞样细胞,在施加外源性bFGF后, SDH在大鼠GMCs中表达升高,并呈现一定的时间、剂量依赖性。 ATG模型中,随着病程的延长,应用western blot方法和免疫组织化学的方法均说明bFGF、 SDH表达升高。结论 bFGF可以诱导SDH的表达升高,而升高的SDH则参与了肾小球肾炎发生发展的过程。

Objective To investigate the influence of the basic fibroblast growth factor ( bFGF) on sorbitol dehydrogenase ( SDH) expression of rat mesangial cells ( MsC) and rat anti Thy-1 glomerular nephritis models. Methods The mesangial cells ( MsC) were cultured by sterile methods. The SDH expression in MsC was detected by western blot methods and meanwhile the expression of bFGF and SDH got detected in ATC. Results MsC became fibroblast- like cells. The SDH expression was higher than normal after exoge-nous bFGF was induced and in a time-dose dependent trend. For the ATC, with the prolongation of disease course, the expression of bFGF and SDH were raised in protein level. Conclusions The bFGF can up-regulate the SDH expression. The SDH can participate in the progress of occurrence and developments of Thy-1 glomerular nephritis.
