目的:探讨Ionomycin诱导精子顶体反应的效果.方法:收集小鼠附睾尾精子,体外获能后用不同终浓度的Ionomycin处理相同时间,以及用相同浓度的Ionomycin处理不同时间.FITC-PNA染色观察并统计精子顶体反应率.结果:用终浓度为1、5、10、50μmol/L Ionomycin 处理1 h 的精子 AR 率分别为31.6%、52.2%、57.4%和64.0%.用终浓度为5μmol/L 的Ionomycin处理10、30、45、60、120、180 min的精子AR率分别为17.2%、25.0%、41.2%、52.2%、54.0%和55.0%.结论:Ionomycin可以诱导小鼠精子的顶体反应,且顶体反应率随着浓度和诱导时间的变化而变化.

Aim:To investigate the spermmatozoa acrosome-reaction (AR)induced by ionomycin. Methods:Motile spermatozoa of the mouse cauda epididymides were treated with ionomycin at different concentrations for different times.Sperm acrosomal status was detected by PNA-FITC staining.Results:Spermatozoa acrosome-reaction was observed after ionomycin treatment.The percentages of acrosome-re-acted spermatozoa were 31.6%,52.2%,57.4%and 64.0%after 1 hour-induction by ionomycin at the final concentration of 1 ,5,1 0,50 μmol/L respectively.When the spermatozoa were treated with 5 μmol/L.Ionomycin for 1 0,30,45,60,1 20,1 80 minutes,the percentages of AR were 1 7.2%,25.0%, 41.2%,52.2%,54.0%,55.0% respectively.Conclusion:Mouse sperm AR has been induced by ionomycin treatment.the percentage of AR sperm varied with the concentration of ionomycin and incuba-tion time.
