为了实现对体外培养细胞生长状态的实时监测,本实验研制出一套可在显微镜下实现细胞实时连续监测的可持续灌注培养系统。本可持续灌注培养系统主要包括4个部分:pH-CO2调控系统、温度控制系统、可持续灌注系统和可拆卸式细胞培养池。可拆卸式细胞培养池由Pro-E软件设计,并以有机玻璃为材料加工。利用此培养系统,已成功地将中国仓鼠卵巢细胞(CHO)、小鼠胚胎成纤维细胞(3 T3)等细胞分别培养4~5天,不但细胞生长状态良好,还可对之进行显微镜下各种形态结构参数的检测。表明该系统可针对某一特定细胞的生命活动进行长时间连续实时追踪测控;并且可与显微操作系统联用,实现单个细胞的搬运,注射等显微操作。

위료실현대체외배양세포생장상태적실시감측,본실험연제출일투가재현미경하실현세포실시련속감측적가지속관주배양계통。본가지속관주배양계통주요포괄4개부분:pH-CO2조공계통、온도공제계통、가지속관주계통화가탁사식세포배양지。가탁사식세포배양지유Pro-E연건설계,병이유궤파리위재료가공。이용차배양계통,이성공지장중국창서란소세포(CHO)、소서배태성섬유세포(3 T3)등세포분별배양4~5천,불단세포생장상태량호,환가대지진행현미경하각충형태결구삼수적검측。표명해계통가침대모일특정세포적생명활동진행장시간련속실시추종측공;병차가여현미조작계통련용,실현단개세포적반운,주사등현미조작。
Inordertomonitorthegrowthstatesofbiologicalcellsinrealtime,acontinuousperfusion culturing system which can realize real-time continuous monitoring of the cells in vitro was developed. The system mainly includes four parts:a pH-CO2 control sub-system,a temperature control sub-system, a continuous perfusion sub-system and a detachable cell culture pool.The detachable cell culture pool was designed by pro-E software and made with PMMA material.CHO and 3 T3 cells were cultured in the cell culture pool for three days.The cells grew well and their sizes,movement and other parameters were detected in real time.Therefore,the continuous perfusion culture system can be used as a monitor for long-term and real-time experiments on cells.By combining with microscopic and micro-manipulation sys-tem,it can be also used to handle and inject cells to perform cell transfection and transferring experi-ments.
