为建立百合高效的离体再生体系,比较了“OT”杂种系百合‘Bay watch’的鳞片、花器官作为外植体时的不定芽诱导和植株再生能力,综合5种外植体分化再生的结果,选择花丝作为最佳外植体材料,结果表明,最适培养基为MS+0.5 mg· L-1 NAA+0.5 mg· L-16-BA+0.2 mg· L-12,4-D,不定芽诱导率高达88.89%。采用两种方式对组培苗进行继代培养,筛选出了直接诱导小鳞茎的适宜培养基配方为MS+(0.1~0.5)mg· L-1 NAA+1.0 mg· L-16-BA;诱导小鳞片分化不定芽的适宜培养基为MS+0.2 mg· L-1 NAA+0.005 mg· L-1 TDZ;诱导生根过程中,适宜的生根培养基为1/2 MS+0.1 mg· L-1 IBA。

We studied the different shoots and plant regeneration ability of Lilium‘Bay watch’ from the bulbscales and different parts of floral organ to establish an efficient regeneration system for Lilium.The filament is the most appropriate among all kinds of explants, andthe optimal medium for shoot regeneration of filament is MS medium supplemented with 0.5 mg· L-1 NAA, 0.5 mg· L-1 6-BA and 0.2 mg· L-1 2,4-D in which the filament has the highest induction rate of 88.89%.Dur-ing the secondary culture, two different reatments were conducted on the test-tube plantlets.The optimal medium for little bulb inducement from thetest -tube plantlets is MSmedium supplemented with 01.-0 .5 mg· L-1 NAA and1 .0 mg· L-1 6-BA.The suitablemedium for shoots proliferation from the bulb scale of the test-tube plantlets is MS medium supplemented with 0.2 mg· L-1 NAA and 0.005 mg· L-1 TDZ.It is beneficial for root growth with 0.1 mg· L-1 IBA in 1/2 MS medium.
