制备了一种注射型壳聚糖(CS)温敏相变复合材料,首先采用乳化-化学交联法,以戊二醛为交联剂,制备包载甲氨蝶呤(MTX)的壳聚糖微球,然后采用离子交联法,向 CS 溶液中滴加50%甘油磷酸钠溶液,制备CS温敏水凝胶.将CS载药微球分散于水凝胶中,制得温敏相变复合材料.结果表明,所制得的微球载药量为12.98%,包封率为32.66%,且粒径均一,分散性良好,具有良好的通针性;水凝胶在37℃下1 min内可发生相变,由溶胶转变为凝胶,具有良好的温度敏感性;制备的温敏相变复合材料具有良好的原位注射性,且4 h 内药物释放率为35.65%,缓释效果明显.

In this article,the inj ectable thermo-sensitive chitosan-based complexity was prepared.First,Metho-trexate (MTX)-loaded chitosan microspheres were prepared with emulsion-chemical cross-linking method using glutaraldehyde as crosslinking agents.CS thermo-sensitive hydrogel was prepared by CS and gycerophosphate using ionic crosslinking method.The thermo-sensitive chitosan-based complexity was composed by CS micro-spheres and CS hydrogel.The results indicated that the drug-loading efficiency and encapsulation efficiency of CS microspheres were 12.98% and 32.66%,respectively.The microspheres prepared in this study had uniform particle size,good dispersibility and could be inj ected through needles.The phase could change from the sol state to a gel state at 3 7 ℃ in 1 min,which indicated the hydrogel prepared in this article had good thermo-sen-sitivity.The thermo-sensitive chitosan-based complexity had a good ability of inj ection.In vitro releasing prop-erties of MTX were investigated and the results indicated that 3 5 .6 5% of the total MTX in the thermo-sensitive chitosan-based complexity was released in the first 4 h.The complexity had an ideal releasing effect.
