目的:探讨阿司匹林对小鼠胚胎腭裂发生的作用,确立阿司匹林诱导小鼠腭裂模型的最佳剂量,为深入研究腭裂提供动物实验模型。方法分别将确定孕期的BALB/C雌鼠随机分为5组,即对照组和实验Ⅰ~Ⅳ组。胚胎第10~12天(embryo 10~embryo 12,E10~ E12)时,每日定时给予对照组孕鼠灌胃生理盐水,实验Ⅰ、Ⅱ、Ⅲ、Ⅳ组孕鼠分别灌胃阿司匹林180、210、240、270 mg/kg。于E18时处死孕鼠,称量并计算孕鼠和胚胎的体质量,记录胚胎腭裂数及发育情况。结果对照组腭裂发生率明显低于实验Ⅲ、Ⅳ组,差异有统计学意义(χ2=15.057, P <00.1;χ2=175.51,P <00.1)。实验Ⅲ、Ⅳ组比较,实验Ⅲ组腭裂发生率高于实验Ⅳ组,差异有统计学意义(χ2=47.42,P <00.1);各组间胚胎体质量的差别无统计学意义(P >00.5)。结论阿司匹林可致小鼠胚胎腭裂发生,240 mg/kg阿司匹林是建立BALB/C胎鼠腭裂模型的最佳剂量。

목적:탐토아사필림대소서배태악렬발생적작용,학립아사필림유도소서악렬모형적최가제량,위심입연구악렬제공동물실험모형。방법분별장학정잉기적BALB/C자서수궤분위5조,즉대조조화실험Ⅰ~Ⅳ조。배태제10~12천(embryo 10~embryo 12,E10~ E12)시,매일정시급여대조조잉서관위생리염수,실험Ⅰ、Ⅱ、Ⅲ、Ⅳ조잉서분별관위아사필림180、210、240、270 mg/kg。우E18시처사잉서,칭량병계산잉서화배태적체질량,기록배태악렬수급발육정황。결과대조조악렬발생솔명현저우실험Ⅲ、Ⅳ조,차이유통계학의의(χ2=15.057, P <00.1;χ2=175.51,P <00.1)。실험Ⅲ、Ⅳ조비교,실험Ⅲ조악렬발생솔고우실험Ⅳ조,차이유통계학의의(χ2=47.42,P <00.1);각조간배태체질량적차별무통계학의의(P >00.5)。결론아사필림가치소서배태악렬발생,240 mg/kg아사필림시건립BALB/C태서악렬모형적최가제량。
Objective To define the optimal dose of aspirin induced cleft palate in BALB/C mice and to pro-vide a reliable model of cleft palate for further study .Methods Pregnant mice at specific period were divid-ed into five groups randomly :experimental groups of Ⅰ ,Ⅱ ,Ⅲ ,Ⅳand control group .The experimental groups were perfued with gastric 180 ,210 ,240 ,270 mg/kg BW ,while the control group only physiologi-cal saline respectively on embryo 10 to embryo 12 .Pregnant mice were killed on embryo 18 to weigh preg-nant mice and embryos and record the number of embryonic development and cleft palate .Results Weight of pregnant mice was significantly different (χ2 = 15 0.57 ,P < 0 0.1;χ2 = 17 5.51 ,P <0 .01) .Incidence of embryonic cleft palate had significant difference between control group and experimental Ⅲ ,Ⅳgroups ,the control group was lower than that in group experiment Ⅲ ,Ⅳ(χ2 =4 7.42 ,P <0 0.1) .The difference was statistically significant that the incidence of cleft palate in experimental group Ⅲ was higher than the ex-perimental group Ⅳ .There was no difference in embryonic weight among each group (P >0 0.5) .Conclu-sion 240 mg/kg is the optimal dose of aspirin to induce cleft palate in BALB/C mice .
