2011和2012年福建省各养殖区引进的美洲鳗鲡( Anguilla Rostrata)和双色鳗鲡( Anguilla bicolor bicolor)在玻璃鳗培育阶段普遍发生“红肝病”,导致了较高的发病率和死亡率,给养殖生产造成严重的危害。本文应用透射电镜( TEM)对各养殖区患“红肝病”的美洲鳗鲡和双色鳗鲡进行超微病理研究。结果显示:在患病美洲鳗鲡鳃组织的肌肉细胞间质中的纤维母细胞查见胞核内许多排列无序的疱疹病毒核衣壳,多数呈空心双环状,直径约80 nm~100 nm,少数核心致密呈线点状或蝌蚪状。在患病双色鳗鲡鳃上皮细胞核旁胞浆内可查见个别成熟病毒粒子。根据所检出病毒的形态和形态发生学特征可以确定该病毒为疱疹病毒科( Herpesviridae)的成员。另外,在病鳗肾细胞的细胞突起之间和细胞外可查见许多大小不等、形态多样,呈杆状、球形、哑铃形或橄榄形,有双层脂质外膜,无细胞壁的支原体( pleuro-pneumonia-like organism,PPLO)。本研究在国内首次应用TEM显微镜意外地在自然感染的养殖鳗鲡组织细胞中检出疱疹病毒和支原体,为鳗鲡“红肝病”的防控提供了有价值的科学依据。

During summer and autumn of 2011and 2012 in Fujian,China,the“red liver disease” with high mortality wide spread over the introduced cultured eels of Anguilla. Rostrata and Anguilla. bicolor bicolor in the glass eel stage and caused serious damage to eel culture. The ultrapathological study of the diseased ells was performed. Under transmission electron microscope, coinfection of herpesvirus and PPLO were detected expectedly. Most of the nucleocapsid of herpesvirus in nucleus of gill fibroblast of infected eel ( A. Rostrata) were round in shape,double-shell with an electron-lucent core,about 80 nm to 100 nm in diameter, and only a few of them with a dense core in the shape of line dot or tadpole. In the cytoplasm of gill epithelial cells of Anguilla bicolor, a few number of virion were noticed. The morphological and morphogenetic characteristics of the virus detected from the infected cultured eels are compatible with the morphology of the family of Herpesviridae. PPLO with polymorphism and variety in size were detected in the ultrasection of the kidney cells of infected cultured eel. The natural detection of coinfection herpesvirus and PPLO in the cultured eels by using electron microscope is the first time in china which provided valuable scientific basis for prevention and control of“red liver disease” in cultured eels.
