为给楠木及其近缘属植物的 DNA 分子鉴定提供依据,对30种楠木及其近缘属植物进行基因组 DNA 提取,叶绿体 matK 序列扩增与测定,并进行序列比对与人工校正,计算属间及种间的遗传距离,比较序列间的差异,构建系统发育树。结果表明:matK 各基因序列的组成中 G+C 含量平均为36.6%。30个楠木及近缘属植物共发现72个碱基变异位点,碱基变异率为9.38%。其 matK 基因序列种间遗传距离为0~0.098,平均遗传距离为0.010。聚类分析,新樟属为1个分支,楠属和润楠属聚为1个分支,其余樟属、木姜子属、新木姜子属、山胡椒属、檫木属、月桂属与黄肉楠属聚为1个分支。故 matK 序列基本可以对楠木及其近缘属植物进行属间鉴定,但在种间水平上并不可行。

To provide references for DNA identification of Phoebe and its related genera,genomic DNA was extracted from 30 accessions,the chloroplast matK sequences were analyzed,the inter-specific genetic distances were calculated,and the phylogenetic trees of all the accessions tested were constructed based on the obtained distance data.The results indicated that the average G+C content in various matK sequences was 36.6%.72 base variation points were found,accounting for 9.38%.The interspecific genetic distance ranged from 0 to 0.098,with an average of 0.010.The cluster analysis showed that the genera tested were clustered into three branches,one was Neocinnamomum delavayi ,one was Phoebe and Machilus,and others belonged to another one,including Cinnamomum,Litsea,Neolitsea,Lindera,Sassafras,Laurus and Actinodaphne.matK could be used to identify Phoebe and its related genera,but couldn't be used at the species level.
