目的探讨 microRNA-21对胆管癌 QBC939细胞株侵袭转移能力的作用。方法构建合成无关序列、microR-NA-21 mimics 和 microRNA-21 inhibitor 并转染至胆管癌QBC939细胞中。实验设自然生长组( Cell )、转染无关序列组( Cell-NC)、转染 microRNA-21 mimics 组( Cell-21M)及转染microRNA-21 inhibitor组( Cell-21I)。运用实时定量 RT-PCR检测各组细胞microRNA-21的表达;通过黏附实验检测各组细胞聚集能力的变化;划痕实验、Transwell迁移观察细胞迁移能力变化, Transwell侵袭实验观察细胞侵袭能力变化。结果与Cell组相比, microRNA-21在 Cell-NC组表达无明显差异,在Cell-21M组表达增加,在Cell-21I组表达降低。黏附实验显示:与Cell组相比,Cel-NC组细胞聚集能力无明显变化,Cell-21M组细胞聚集能力增强,Cell-21I组细胞聚集能力减弱(P<0.05);划痕实验及Transwell迁移实验示Cell-NC组细胞迁移能力无明显变化,Cell-21M组细胞迁移能力增强,Cell-21I细胞聚集能力减弱(P<0.05);Transwell侵袭实验示Cell-NC组细胞侵袭能力无明显变化,Cell-21M组细胞侵袭能力增强, Cell-21I 细胞侵袭能力减弱( P <0.05)。结论microRNA-21可增强胆管癌QBC939细胞的侵袭与转移能力,提示microRNA鄄21可能在胆管癌的侵袭转移过程中发挥重要作用。

목적탐토 microRNA-21대담관암 QBC939세포주침습전이능력적작용。방법구건합성무관서렬、microR-NA-21 mimics 화 microRNA-21 inhibitor 병전염지담관암QBC939세포중。실험설자연생장조( Cell )、전염무관서렬조( Cell-NC)、전염 microRNA-21 mimics 조( Cell-21M)급전염microRNA-21 inhibitor조( Cell-21I)。운용실시정량 RT-PCR검측각조세포microRNA-21적표체;통과점부실험검측각조세포취집능력적변화;화흔실험、Transwell천이관찰세포천이능력변화, Transwell침습실험관찰세포침습능력변화。결과여Cell조상비, microRNA-21재 Cell-NC조표체무명현차이,재Cell-21M조표체증가,재Cell-21I조표체강저。점부실험현시:여Cell조상비,Cel-NC조세포취집능력무명현변화,Cell-21M조세포취집능력증강,Cell-21I조세포취집능력감약(P<0.05);화흔실험급Transwell천이실험시Cell-NC조세포천이능력무명현변화,Cell-21M조세포천이능력증강,Cell-21I세포취집능력감약(P<0.05);Transwell침습실험시Cell-NC조세포침습능력무명현변화,Cell-21M조세포침습능력증강, Cell-21I 세포침습능력감약( P <0.05)。결론microRNA-21가증강담관암QBC939세포적침습여전이능력,제시microRNA견21가능재담관암적침습전이과정중발휘중요작용。
Objective To detect the effects of microRNA-21 on invasion and metastasis of cholangiocarcinoma QBC939 cells. Methods MicroRNA-21 scamble, mimics and inhibitor were constructed and transfected into QBC939 cells. The blank control gruop( Cell) ,negative control group( Cell-NC) ,micorRNA-21 mimics group( Cell-21M)and microRNA-21 inhibitor group(Cell-21I)were designed in this study. The expression of microRNA-21 was examined by real-time RT-PCR. Then adhesion experiment was used to examin aggregation capability. Wound healing experiment and transwell migration experiment were used to investigate the the migration capability. Tran-swell invasion experiment was used to investigate the invasion capability of cells in each group. Results Compa-ring with Cell, the expression of microRNA-21 in Cell-21M was added and in Cell-21I was reduced. Adhesion ex-periment showed that aggregation capability of cells in Cell-21 M was increased and in Cell-21 I decreased ( P <0.05 ) . Wound healing experiment and transwell migration experiment showed that migration capability of cells in Cell-21 M was increased and in Cell-21 I decreased ( P<0.05 ) . Transwell invasion experiment showed that invasion capability of cells in Cell-21 M was increased and in Cell-21 I decreased ( P<0.05 ) . All the results had no differ-ent significance in Cell-NC. Conclusion MicroRNA-21 can promote the invasion and metastasis of cholangiocarci-noma QBC939 cells, which suggest that microRNA-21 may play a vital role in the process of invasion and metastasis of cholangiocarcinoma.
