The level of free proline content in plant tissues is often regarded as a physiological index indicating the strength of plant anti-adversity. This paper explores and analyzes the reasons and elimination methods of the extraction and emulsification phe-nomenon of toluene in the process of determining the index by acidic ninhydrin colorimetry method, hoping to provide refer-ences for scientific researchers and students engaging in the de-termination of the index.
