[目的]通过建立帕金森病大鼠模型,观察头穴电针对帕金森病大鼠行为学的影响,同时检测帕金森病大鼠黑质中脑源性神经营养因子(Brain-derived neurotrophic factor,BDNF)及其受体酪氨酸激酶B(Tyrosine kinase-B,TrkB),以及黑质中cAMP反应元件结合蛋白(cAMP respon-sive element binding protein,CREB)的变化,以探讨针灸治疗帕金森病的可能机理。[方法]用纹状体内多位点注射6-OHDA建立帕金森病大鼠模型,造模成功者随机分为电针治疗组、模型对照组、假手术组和正常组,分别进行头穴电针治疗。疗程结束后采用免疫组化法检测黑质中BDNF、TrkB、CREB水平。[结果]头穴电针治疗组大鼠治疗后较治疗前旋转次数明显减少(P<0.05),模型对照组和电针治疗组黑质内BDNF、TrkB和CREB阳性神经元数量显著低于正常组和假手术组(P<0.01),电针治疗组BDNF、TrkB和CREB阳性神经元数量高于模型对照组(P<0.05)。[结论]头穴电针可明显改善帕金森大鼠的旋转行为,增加黑质中BDNF、TrkB和CREB的阳性细胞数。其机制可能为通过增加黑质中BDNF和TrkB的含量,激活神经元信号保护通路从而使CREB活化,促进了BDNF在黑质内的自分泌,保护DA能神经元。

[목적]통과건립파금삼병대서모형,관찰두혈전침대파금삼병대서행위학적영향,동시검측파금삼병대서흑질중뇌원성신경영양인자(Brain-derived neurotrophic factor,BDNF)급기수체락안산격매B(Tyrosine kinase-B,TrkB),이급흑질중cAMP반응원건결합단백(cAMP respon-sive element binding protein,CREB)적변화,이탐토침구치료파금삼병적가능궤리。[방법]용문상체내다위점주사6-OHDA건립파금삼병대서모형,조모성공자수궤분위전침치료조、모형대조조、가수술조화정상조,분별진행두혈전침치료。료정결속후채용면역조화법검측흑질중BDNF、TrkB、CREB수평。[결과]두혈전침치료조대서치료후교치료전선전차수명현감소(P<0.05),모형대조조화전침치료조흑질내BDNF、TrkB화CREB양성신경원수량현저저우정상조화가수술조(P<0.01),전침치료조BDNF、TrkB화CREB양성신경원수량고우모형대조조(P<0.05)。[결론]두혈전침가명현개선파금삼대서적선전행위,증가흑질중BDNF、TrkB화CREB적양성세포수。기궤제가능위통과증가흑질중BDNF화TrkB적함량,격활신경원신호보호통로종이사CREB활화,촉진료BDNF재흑질내적자분비,보호DA능신경원。
[Objective] The experiment wil study the effect of the electroacupuncture(EA) on PD rats that of Brain-derived neurotrophic factor(BDNF), tyrosine kinase-B(TrkB) and cAMP responsive element binding protein(CREB) in order to know the possible mechanisms. [Methods]The research created rats model of PD was caused by injecting 6-hydroxydopamine into the left striatum from 4 points use stereotaxic apparatus. Then histochemical and im-munohistochemical staining methods were adopted to observe the therapy effects of the BDNF, TrkB and CREB content in substantia nigra. [Results](1) In EA group, the number of rotations after the treatment was lower than before the treatment(P<0.05), but that didn't change in PD group(P>0.05).(2) The number of BDNF, TrkB and CREB positive neurons in PD group was obviously less than those of the normal group and control group( P<0.01).In the EA group, the number of BDNF, TrkB and CREB positive neurons was more than that of the model group( P<0.05).[Conclusion]EA on head can increase the number of BDNF, TrkB and CREB in substantia nigra, thus enhances BDNF in the substantia nigra from the secretion and protects DA neurons.
