目的:建立高效液相色谱-蒸发光检测法测定白头翁皂苷D含量的方法。方法 Hyperisil ODS2 C18柱(250 mm ×4.6 mm ,5μm),柱温30℃;流动相:甲醇-水-甲酸(72∶28∶0.1);流速1 mL · min-1;Altech ELSD 3300检测器,雾化气流速1.3 L · min-1,漂移管温度为55℃。结果该方法的线性范围为0.1046~1.0460 mg · mL -1(r=0.9992),平均回收率(n=6)为101.0%(RSD=1.6%),不同产地药材白头翁皂苷D差异性较大。结论该方法专属性强、准确、灵敏度高、重复性好,可用于白头翁中白头翁皂苷D的测定,对白头翁规范性种植具有指导意义。

목적:건립고효액상색보-증발광검측법측정백두옹조감D함량적방법。방법 Hyperisil ODS2 C18주(250 mm ×4.6 mm ,5μm),주온30℃;류동상:갑순-수-갑산(72∶28∶0.1);류속1 mL · min-1;Altech ELSD 3300검측기,무화기류속1.3 L · min-1,표이관온도위55℃。결과해방법적선성범위위0.1046~1.0460 mg · mL -1(r=0.9992),평균회수솔(n=6)위101.0%(RSD=1.6%),불동산지약재백두옹조감D차이성교대。결론해방법전속성강、준학、령민도고、중복성호,가용우백두옹중백두옹조감D적측정,대백두옹규범성충식구유지도의의。
Objective To establish a determination method of pulsatilla saponin D in Pulsatilla chinensis by HPLC-ELSD method . Methods Samples were analyzed on a Hyperisil ODS2 C18 column(250 mm × 4 .6 mm ,5μm) and methanol-water-formic acid(72∶28∶0 .1) was used as the mobile phase .The flow rate was 1 mL · min-1 and the temperature was 30 ℃ .The Altech ELSD 3300 detector was uesd as a detector .The flow rate of atomizer was 1 .3 L · min-1 and the temperature of drift tube was 55 ℃ .Result The method had a linear relationship in the range of 0 .104 6-1 .046 0 mg · mL -1 (r=0 .999 2) and the average recovery rate was 101.0% (n=6 ,RSD 1 .6% ) .The contents of pulsatilla saponin D were much different in different areas .Conclusion The method has advantages of high specific ,accurate ,sensitive and reproducible ,and can be used for the determination of pulsatilla saponin D in Pulsatilla chinensis ,which has a pioneer significance for Pulsatilla chinensis standard planting .
