Oxytropis falcata%total alkaloids%extraction and purification
目的:研究藏药镰形棘豆总生物碱的提取纯化的最佳工艺。方法选择物料比、乙醇体积分数、醋酸体积分数和提取温度作为实验的考察因素,以酸性染料比色法测定吸光度为指标,采用正交实验优选镰形棘豆总生物碱的提取纯化最佳工艺。结果最优工艺条件为以5mL·L-1醋酸的体积分数40%乙醇溶液为溶剂,物料比为1∶20,60℃超声提取2次,经732型阳离子交换树脂进行吸附,再以10mL·L-1NH3·H2O的体积分数60%乙醇溶液超声提取树脂中的生物碱。结论优选的工艺稳定可行,能较好地提取镰形棘豆中的总生物碱,为今后从镰形棘豆中获得总生物碱提供了科学依据。
目的:研究藏藥鐮形棘豆總生物堿的提取純化的最佳工藝。方法選擇物料比、乙醇體積分數、醋痠體積分數和提取溫度作為實驗的攷察因素,以痠性染料比色法測定吸光度為指標,採用正交實驗優選鐮形棘豆總生物堿的提取純化最佳工藝。結果最優工藝條件為以5mL·L-1醋痠的體積分數40%乙醇溶液為溶劑,物料比為1∶20,60℃超聲提取2次,經732型暘離子交換樹脂進行吸附,再以10mL·L-1NH3·H2O的體積分數60%乙醇溶液超聲提取樹脂中的生物堿。結論優選的工藝穩定可行,能較好地提取鐮形棘豆中的總生物堿,為今後從鐮形棘豆中穫得總生物堿提供瞭科學依據。
목적:연구장약렴형극두총생물감적제취순화적최가공예。방법선택물료비、을순체적분수、작산체적분수화제취온도작위실험적고찰인소,이산성염료비색법측정흡광도위지표,채용정교실험우선렴형극두총생물감적제취순화최가공예。결과최우공예조건위이5mL·L-1작산적체적분수40%을순용액위용제,물료비위1∶20,60℃초성제취2차,경732형양리자교환수지진행흡부,재이10mL·L-1NH3·H2O적체적분수60%을순용액초성제취수지중적생물감。결론우선적공예은정가행,능교호지제취렴형극두중적총생물감,위금후종렴형극두중획득총생물감제공료과학의거。
Objective To optimize the conditions for extraction and purification of total alkaloids from Tibetan medicine ,Oxytropis f alcata .Methods On the basis of single factor investigation ,four factors were chosen to optimize the preparing conditions by an orthogonal design ,i .e .the ratio of material to reagent ,the temperature of extraction ,the concentrations of ethanol and HAc . Absorbance determined by acidic dye colorimetry was adopted as index . Results The optimal conditions were as follows :extracting twice aided by ultrasonic with 40% ethanol solution containing 5 mL · L -1 HAc at 60 ℃ ,the ratio of material to reagent 1∶20 ,purifying by 732 cation exchange resin and then extracting with 60% ethanol solution containing 10 mL · L -1 NH3 · H2 O .Conclusion The optimal method is stable and reproducible and can be used for the extraction and purification of total alkaloids from Tibetan medicine O . f alcata .
