目的:建立并验证后天感染河南樱桃谷鸭乙型肝炎病毒( DHBV)模型。方法:筛选3日龄DHBV阴性河南樱桃谷鸭,经胫静脉注射DHBV阳性血清建立DHBV模型鸭,连续观察42 d,采用荧光定量PCR方法检测鸭血清和肝脏中DHBV DNA变化情况。 DHBV阴性鸭48只,分为正常组、模型组和3TC组,每组16只。模型组和3TC组于鸭3日龄时建模,模型组给予生理盐水2 mL/(kg· d),3TC组给予3TC 20 mg/(kg· d),连续给药10 d。 PCR检测给药5、10 d后及停药3 d后血清和肝脏中DHBV DNA含量,并进行肝脏病理学观察。结果:感染2 d后,鸭血清和肝脏中均可检测到DHBV DNA。血清DHBV DNA水平第14天达到高峰,拷贝数为7.7×109 mL-1,肝脏DHBV DNA第21天达到高峰,拷贝数达2.1×109 mL-1,42 d内均维持较高的DHBV DNA水平。与模型组比较,3TC组鸭血清和肝脏DHBV DNA拷贝数明显降低(血清:F组间=17.625,F时间=68.141,F交互=134.517,P<0.001;肝脏:F组间=69.430,F时间=35.330,F交互=75.770,P<0.001)。3TC组鸭用药10 d后,血清和肝脏HBV DNA抑制率分别为86.5%和84.9%,但停药3 d后有轻度“反弹”现象。3TC组肝组织病变较模型组有明显改善。结论:河南樱桃谷鸭后天感染DHBV较敏感,病毒在体内增殖稳定,是DHBV感染的理想动物模型。

목적:건립병험증후천감염하남앵도곡압을형간염병독( DHBV)모형。방법:사선3일령DHBV음성하남앵도곡압,경경정맥주사DHBV양성혈청건립DHBV모형압,련속관찰42 d,채용형광정량PCR방법검측압혈청화간장중DHBV DNA변화정황。 DHBV음성압48지,분위정상조、모형조화3TC조,매조16지。모형조화3TC조우압3일령시건모,모형조급여생리염수2 mL/(kg· d),3TC조급여3TC 20 mg/(kg· d),련속급약10 d。 PCR검측급약5、10 d후급정약3 d후혈청화간장중DHBV DNA함량,병진행간장병이학관찰。결과:감염2 d후,압혈청화간장중균가검측도DHBV DNA。혈청DHBV DNA수평제14천체도고봉,고패수위7.7×109 mL-1,간장DHBV DNA제21천체도고봉,고패수체2.1×109 mL-1,42 d내균유지교고적DHBV DNA수평。여모형조비교,3TC조압혈청화간장DHBV DNA고패수명현강저(혈청:F조간=17.625,F시간=68.141,F교호=134.517,P<0.001;간장:F조간=69.430,F시간=35.330,F교호=75.770,P<0.001)。3TC조압용약10 d후,혈청화간장HBV DNA억제솔분별위86.5%화84.9%,단정약3 d후유경도“반탄”현상。3TC조간조직병변교모형조유명현개선。결론:하남앵도곡압후천감염DHBV교민감,병독재체내증식은정,시DHBV감염적이상동물모형。
Aim:To establish a standardized duck hepatitis B virus ( DHBV) animal model using Cherry Valley ducks of Henan Province .Methods:3-day-old DHBV negative ducks were selected and injected DHBV positive serum via tibial vein.Fluorescent quantitative polymerase chain reaction ( FQ-PCR) was used to detect the levels of DHBV DNA in serum and liver for 42 days.A total of 48 DHBV negative ducks were allocated into 3 groups:the model group and lamivudine (3TC) group were established DHBV model and given normal saline at 2 mL/(kg· d) and 3TC at 20 mg/(kg· d) for 10 days, respectively.DHBV DNA in the serum and liver were detected after medicating for 5 days, 10 days, and after with-drawal of the drug for 3 days.Histopathological examination of duck liver was also performed .Results:DHBV DNA in ser-um and liver could be detected on the 2nd day of post-infection.The level of DHBV DNA in serum increased to a peak at Day 14 with 7.7 ×109 copies/mL.The level of DHBV DNA in liver increased to a peak at Day 21 with 2.1 ×109 copies/mL.DH-BV DNA maintained a high level in 42 days.3TC [20 mg/(kg· d)] had a strong inhibitory effect on DHBV DNA .The in-hibition rate reached 86.5%and 84.9%in duck serum and liver on Day 10.The histopathological evaluation of the liver also revealed significant improvement .However, a rebound of DHBV DNA level was observed at Day 3 post-treatment.Conclu-sion:Henan Cherry Valley ducks are sensitive to DHBV and the level of virus in the serum and liver is stable , making it an ideal disease model for DHBV infection and can be used for the screening and evaluation of anti -HBV drugs.
