目的:检测非小细胞肺癌( NSCLC)组织中Nanog的表达。方法:采用免疫组化及实时荧光定量PCR法分别检测106例NSCLC及10例正常肺组织中Nanog蛋白及mRNA的表达,采用生存分析探讨Nanog蛋白的表达与肺癌患者预后的关系。结果:NSCLC组织中Nanog蛋白的阳性表达率为24.5%(26/106),高于正常肺组织(0%,P=0.114);Nanog蛋白在肺腺癌中的表达明显高于肺鳞状细胞癌(肺鳞癌)(χ2=5.087,P=0.024),随病理级别增加, Nanog蛋白的阳性表达率升高(χ2=6.845,P=0.033);Nanog蛋白的表达与肺鳞癌患者的预后无关(χ2=0.013,P=0.917),肺腺癌患者中Nanog阴性表达者预后较好(χ2=5.352,P=0.020)。 NSCLC组织中Nanog mRNA 的表达为(0.950±0.183),高于正常肺组织的(0.277±0.190)(F=122.662,P<0.001);随病理级别增加,Nanog mRNA的表达升高(F=4.047,P=0.020)。结论:Nanog的表达与NSCLC的发生发展有关,且Nanog蛋白的表达与肺腺癌的预后有关。

목적:검측비소세포폐암( NSCLC)조직중Nanog적표체。방법:채용면역조화급실시형광정량PCR법분별검측106례NSCLC급10례정상폐조직중Nanog단백급mRNA적표체,채용생존분석탐토Nanog단백적표체여폐암환자예후적관계。결과:NSCLC조직중Nanog단백적양성표체솔위24.5%(26/106),고우정상폐조직(0%,P=0.114);Nanog단백재폐선암중적표체명현고우폐린상세포암(폐린암)(χ2=5.087,P=0.024),수병리급별증가, Nanog단백적양성표체솔승고(χ2=6.845,P=0.033);Nanog단백적표체여폐린암환자적예후무관(χ2=0.013,P=0.917),폐선암환자중Nanog음성표체자예후교호(χ2=5.352,P=0.020)。 NSCLC조직중Nanog mRNA 적표체위(0.950±0.183),고우정상폐조직적(0.277±0.190)(F=122.662,P<0.001);수병리급별증가,Nanog mRNA적표체승고(F=4.047,P=0.020)。결론:Nanog적표체여NSCLC적발생발전유관,차Nanog단백적표체여폐선암적예후유관。
Aim:To investigate the expression of Nanog in non-small cell lung cancer (NSCLC) tissue.Methods:The expressions of Nanog mRNA and protein were detected in the specimens from 106 cases of NSCLC and 10 cases of normal lung tissue by using quantitative real-time PCR and immunohistochemistry , respectively , then the relationship between the expression of Nanog protein and the prognosis of the patients with lung adenocarcinoma ( LAC) or lung squmous cell carci-noma(LSCC) was analyzed by log-rank test.Results:The positive expression rate of Nanog protein in NSCLC tissue was 24.5%,higher than that(0%) in normal lung tissue(P=0.114);the positive expression rate of Nanog protein was higher in LAC tissue than that in LSCC tissue and increased with the increase of pathological degree (χ2 =5.087,6.845,P <0.05);in patients with LAC, those with negative expression of Nanog protein had better progonosis (χ2 =5.352,P=0 .020).The expression level of Nanog mRNA in NSCLC tissue was (0.950 ±0.183),higher than that (0.277 ±0.190) in normal lung tissue(F=122.662,P<0.001);the expression level of Nanog mRNA increased with the increase of patho-logical degree(F=4.047,P=0.020).Conclusion:Nanog expression is relevant to the carcinogenesis and development of NSCLC, besides,the expression of Nanog protein is associated with the prognosis of patients with LAC .
