近年来发现郑州地区的番茄表现植株矮化、叶片黄化曲叶等类似番茄黄化曲叶病毒病症状,为了确定疑似病毒的分类地位,根据已知的番茄黄化曲叶病毒(TYLCV )基因组序列设计特异性引物TYLCV CPF/TYLC CPR ,通过PCR扩增其外壳蛋白基因,对获得的特异性片段回收、克隆、测序并进行同源性比较及分子系统进化分析。结果显示,郑州分离物的外壳蛋白基因由777个核苷酸组成,其核苷酸和编码氨基酸序列与番茄黄化曲叶病毒其他分离物同源性均在95%以上;系统进化分析表明,郑州分离物与北京、烟台、上海和新疆分离物亲缘关系最近。根据双生病毒的分类标准,判断郑州分离物应属于番茄黄化曲叶病毒的一个分离物。

근년래발현정주지구적번가표현식주왜화、협편황화곡협등유사번가황화곡협병독병증상,위료학정의사병독적분류지위,근거이지적번가황화곡협병독(TYLCV )기인조서렬설계특이성인물TYLCV CPF/TYLC CPR ,통과PCR확증기외각단백기인,대획득적특이성편단회수、극륭、측서병진행동원성비교급분자계통진화분석。결과현시,정주분리물적외각단백기인유777개핵감산조성,기핵감산화편마안기산서렬여번가황화곡협병독기타분리물동원성균재95%이상;계통진화분석표명,정주분리물여북경、연태、상해화신강분리물친연관계최근。근거쌍생병독적분류표준,판단정주분리물응속우번가황화곡협병독적일개분리물。
Yellow leaf curl symptoms have been observed on tomato for the first time in many pre-fectures of Zhengzhou during the past several years .To identify the suspected begomovirus ,a pair of specific primers TYLCV-CPF/TYLCV-CPR was designed according to known genome se-quences of tomato yellow leaf curl virus (TYLCV) to conduct PCR ,and then the generated spe-cific segment was collected ,cloned and sequenced .The homology comparison and phylogenetic analysis of the coat protein gene were made at last .The result indicated that the complete nucleo-tide sequence of the coat protein gene was 777 bp .The homology of the isolate from Zhengzhou was 95% or more with the other TYLCV isolates at nucleotide and amino acid level .The phyloge-netic analysis showed that the Zhengzhou isolate was closely related to Beijing ,Yantai ,Shanghai , Xinjiang isolates of TYLCV .According to the classification criteria of geminivirus ,the Zhengzhou isolate should belong to an isolate of TYLCV .
