目的:研究局部转染Ngn2基因对急性脊髓损伤大鼠运动功能的影响。方法:将36只4月龄SPF级SD大鼠,随机分成3组,分别为A组(假手术组)、B组(实验组)、C组(对照组),每组12只。假手术组只咬除T8~T10棘突及椎板、不损伤脊髓,实验组和对照组采用改良Allen's打击法制造T8急性脊髓损伤模型,术后分别向B、C 2组损伤脊髓节段持续注射Ngn2质粒和空载体质粒各5μg。于术后1、2、3、4周采用BBB运动功能评分系统检测大鼠运动功能,术后1周应用RT-PCR检测损伤节段脊髓组织Ngn2基因表达,术后4周HE染色脊髓病理学检测及SP染色检测Nestin+细胞数。结果:术后各时间点实验组BBB评分明显优于对照组,有统计学意义(P<0.05);实验组大鼠脊髓组织中Ngn2mRNA有表达,而对照组无表达;实验组单个视野Nestin+细胞为(14.25±2.37)个,对照组单个视野Nestin+细胞为(2.54±1.14)个,差异有统计学意义( P<0.05)。结论:Ngn2基因能够促进急性脊髓损伤大鼠运动功能恢复,可能与诱导脊髓内源性神经干细胞的增殖有关。

목적:연구국부전염Ngn2기인대급성척수손상대서운동공능적영향。방법:장36지4월령SPF급SD대서,수궤분성3조,분별위A조(가수술조)、B조(실험조)、C조(대조조),매조12지。가수술조지교제T8~T10극돌급추판、불손상척수,실험조화대조조채용개량Allen's타격법제조T8급성척수손상모형,술후분별향B、C 2조손상척수절단지속주사Ngn2질립화공재체질립각5μg。우술후1、2、3、4주채용BBB운동공능평분계통검측대서운동공능,술후1주응용RT-PCR검측손상절단척수조직Ngn2기인표체,술후4주HE염색척수병이학검측급SP염색검측Nestin+세포수。결과:술후각시간점실험조BBB평분명현우우대조조,유통계학의의(P<0.05);실험조대서척수조직중Ngn2mRNA유표체,이대조조무표체;실험조단개시야Nestin+세포위(14.25±2.37)개,대조조단개시야Nestin+세포위(2.54±1.14)개,차이유통계학의의( P<0.05)。결론:Ngn2기인능구촉진급성척수손상대서운동공능회복,가능여유도척수내원성신경간세포적증식유관。
Objective:To investigate the effect of local Ngn 2 plasmid transfer on motor function in rats with acute spinal cord injury motor function.Methods:36 four-month-old SPF SD rats were randomly divided into three groups ,namely Group A (sham group), Group B (experimental group), Group C (control group),n =12.Sham group was only bite T8-T10 spinous process and lamina with-out damage to the spinal cord , the experimental group and the control group made the T 8 acute spinal cord injury model by using modified Allen's method.Postoperatively, 5μg Ngn2 plasmid for Group B and 5μg vector plasmid for Group C were continuously injected into the spinal cord segments .1,2,3,4 weeks after surgery to detect motor function in rats using the BBB motor function scoring system , 1 week after surgery to test the expression of Ngn 2 gene of spinal cord injury tissue by RT -PCR and 4 weeks after surgery detect spinal pathology by HE staining or the number of Nestin +cells by SP staining .Results:Postoperative BBB score the experimental group at each time point was significantly better than the control group .It was statistically significant (P <0.05).There was the expression of Ngn2 gene in spinal cord injury tissue of the experimental group , no expression in the control group .The experimental group had Nestin +cells (14. 25 ±2.37) ones per a single vision, while the control group Nestin +cells (2.54 ±1.14) ones, the difference was statistically sig-nificant (P <0.05).Conclusion:Ngn2 gene transferred by local injection has significant effect on the motor function recovery in rats with acute spinal cord injury , which may induce proliferation of the endogenous neural stem cells in the spinal cord .
