目的探讨Dishevelled蛋白在人脑胶质瘤中的表达情况,并分析其表达水平与临床病理级别的关系。方法采用免疫组化法及免疫印迹法检测Dishevelled蛋白在67例不同病理级别人脑胶质瘤标本和3例正常人脑组织中的表达水平。结果在正常人脑组织中均未见Dishevelled蛋白表达,而在人脑胶质瘤组织中Dishevelled蛋白高表达,67例胶质瘤中,Dishevelled阳性率为65.67%(44/67);且 Dishevelled蛋白表达水平均随着脑胶质瘤病理级别的增高而显著升高(P<0.05)。结论 Dishevelled蛋白高表达可能对人脑胶质瘤细胞的发生、发展具有促进作用。本研究结果不仅为Wnt/β-catenin信号通路的研究开辟了新的领域,也为诊断及基因治疗脑胶质瘤提供了新的靶点。
目的探討Dishevelled蛋白在人腦膠質瘤中的錶達情況,併分析其錶達水平與臨床病理級彆的關繫。方法採用免疫組化法及免疫印跡法檢測Dishevelled蛋白在67例不同病理級彆人腦膠質瘤標本和3例正常人腦組織中的錶達水平。結果在正常人腦組織中均未見Dishevelled蛋白錶達,而在人腦膠質瘤組織中Dishevelled蛋白高錶達,67例膠質瘤中,Dishevelled暘性率為65.67%(44/67);且 Dishevelled蛋白錶達水平均隨著腦膠質瘤病理級彆的增高而顯著升高(P<0.05)。結論 Dishevelled蛋白高錶達可能對人腦膠質瘤細胞的髮生、髮展具有促進作用。本研究結果不僅為Wnt/β-catenin信號通路的研究開闢瞭新的領域,也為診斷及基因治療腦膠質瘤提供瞭新的靶點。
목적탐토Dishevelled단백재인뇌효질류중적표체정황,병분석기표체수평여림상병리급별적관계。방법채용면역조화법급면역인적법검측Dishevelled단백재67례불동병리급별인뇌효질류표본화3례정상인뇌조직중적표체수평。결과재정상인뇌조직중균미견Dishevelled단백표체,이재인뇌효질류조직중Dishevelled단백고표체,67례효질류중,Dishevelled양성솔위65.67%(44/67);차 Dishevelled단백표체수평균수착뇌효질류병리급별적증고이현저승고(P<0.05)。결론 Dishevelled단백고표체가능대인뇌효질류세포적발생、발전구유촉진작용。본연구결과불부위Wnt/β-catenin신호통로적연구개벽료신적영역,야위진단급기인치료뇌효질류제공료신적파점。
Objective To investigate the expression of Dishevelled protein in the human brain glioma, and analysis of the relationship between its expression and clinical pathological grading.Methods Expression levels of Dishevelled protein were detected by immunohistochemistry and western blot in 67 patients with different pathological grades of human gliomas and 3 normal brain tissues. Results There were no Dishevelled protein expression in normal human brain tissues,and Dishevelled protein overexpressed in human glioma tissue,the positive rate was 65.67% (44/67). The expression level of Dishevelled was increased with the ascending of pathological grade of brain glioma(P<0.05). Conclusion Dishevelled protein overexpression may be closely related with occurrence and development of human brain gliomas. The results of this study not only sets up a new field for the study of Wnt/β -catenin signal pathway, but also provides a new target for the diagnosis and gene therapy of brain gliomas.
