目的:探讨鼻腔局部给予重组小鼠白细胞介素22(IL-22)对小鼠变应性鼻炎免疫应答的调节作用。方法将24只6~8周的雌性BALB/c小鼠随机分为3组,每组8只,分别为对照组、模型组和IL-22组。以卵白蛋白(ovalbumin/OVA)为致敏原,氢氧化铝凝胶为佐剂制备变应性鼻炎小鼠模型,于激发前给予小鼠鼻腔局部滴注重组小鼠IL-22。观察小鼠变应性鼻炎症状改善情况,并检测三组小鼠鼻黏膜局部嗜酸性粒细胞侵袭情况、外周血清中OVA-特异性IgE(OVA-sIgE)浓度及Th1、Th2细胞因子的表达情况。结果重组小鼠IL-22干预的变应性鼻炎小鼠症状较模型组缓解,鼻腔黏膜局部嗜酸性粒细胞侵袭减少, OVA-sIgE浓度降低,Th2型细胞因子IL-4和IL-5浓度下降。差异均具有统计学意义(P<0.05)。Th1型细胞因子IFN-γ浓度在IL-22治疗组和模型组间差异无统计学意义。结论鼻腔局部给予重组小鼠IL-22可以抑制变应性鼻炎小鼠的免疫反应,它有望成为治疗变应性鼻炎的一种新方法。

목적:탐토비강국부급여중조소서백세포개소22(IL-22)대소서변응성비염면역응답적조절작용。방법장24지6~8주적자성BALB/c소서수궤분위3조,매조8지,분별위대조조、모형조화IL-22조。이란백단백(ovalbumin/OVA)위치민원,경양화려응효위좌제제비변응성비염소서모형,우격발전급여소서비강국부적주중조소서IL-22。관찰소서변응성비염증상개선정황,병검측삼조소서비점막국부기산성립세포침습정황、외주혈청중OVA-특이성IgE(OVA-sIgE)농도급Th1、Th2세포인자적표체정황。결과중조소서IL-22간예적변응성비염소서증상교모형조완해,비강점막국부기산성립세포침습감소, OVA-sIgE농도강저,Th2형세포인자IL-4화IL-5농도하강。차이균구유통계학의의(P<0.05)。Th1형세포인자IFN-γ농도재IL-22치료조화모형조간차이무통계학의의。결론비강국부급여중조소서IL-22가이억제변응성비염소서적면역반응,타유망성위치료변응성비염적일충신방법。
Objective Explore regulatory effect of immune response in mice with allergic rhinitis by nasal topical administration of recombinant mouse interleukin 22(IL-22).Methods 6 to 8 weeks,24 female BALB/c mice were randomly divided into three groups,each group of 8,respectively,for control group,the model group and IL-22 treatment group.Allergic mice were prepared with chicken ovalbumin as allergen and aluminium hydroxide gel as immune adjuvant.Recombinant mouse IL-22 was administrated into the nostril.Observe the symptoms of allergic rhinitis mouse and detected nasal mucosa local eosinophil invasion,concentration of peripheral serum ovalbumin-specific IgE(OVA-sIgE) and Th1,Th2 cytokine expression.Results Symptoms of recombinant mouse IL-22 intervention allergic rhinitis mice were relieved compared to the model group.Reduce local eosinophil invasion of the nasal mucosa,concentration of OVA-s IgE decrease and levels of Th2 cytokines IL-4 and IL-5 levels drop down.Differences were statistically significant (P<0.05).The differences of Th1 cytokines IFN-γ concentration between IL-22 group and model group have no statically significance.Conclusion Nasal locally administered recombinant mouse IL-22 can inhibit the allergic reaction in mice.IL-22 is expected to become a new method for treatment of allergic rhinitis.
