利用盐酸多巴酚丁胺对血红蛋白模拟酶催化L-酪氨酸和H2O2体系的抑制作用,建立了酶催化荧光法测定盐酸多巴酚丁胺的新方法。研究了该猝灭反应的最佳实验条件,其猝灭程度与盐酸多巴酚丁胺的浓度呈线性关系。测定的线性范围为5.92×10-7~8.88×10-6mol/L,检出限为1.43×10-9mol/L。对浓度为5.92×10-7mol/L的盐酸多巴酚丁胺溶液进行11次平行测定,其相对标准偏差为3.2%。该方法用于药物中盐酸多巴酚丁胺含量的测定,结果满意。

A high sensitive and simple spectrofluorimetric method for the determination of dobutanmine hydrochloride based on its inhibitory effect on the hemoglobin-catalyzed reaction of H2O2 and L-tyrosine was developed. The concentration of dobutanmine hydrochloride is in linear relationship with the fluorescence quenching (ΔF) of system under the optimal experimental con-ditions. The calibration graph is linear in the range from 5.92í10-7 to 8.88í10-6 mol/L with the detection limit of 1.43í10-9 mol/L. The relative standard deviation for 11 replicate determi-nation of 5.92í10-7 mol/L dobutamine hydrochloride was 3.2%. This method can be used for the determination of dobutanmine hydrochloride in its pharmaceutical formulations and urine with satisfactory results.
